肝劳发生机理的实验研究之二

【摘要】 目的:从支配瞳孔括约肌的副交感神经系统的神经-受体-第二信使以及瞳孔括约肌的病理组织学角度探讨肝劳的发生机理。 方法:以树鼩作为实验动物,采用限定照明时间并予以形觉刺激诱视的方法,迫使树鼩近距离久视以模拟肝劳的主要发病原因——“过用目力”。在实验的第3、6、9天分别处死模型组及正常对照组动物,取虹膜检测其MAChR(muscarinic acetylcholine receptor毒蕈碱乙酰胆碱受体,简称M受体)的RT值(受体最大结合容量)和K_D值(平衡解离常数),以观察造模3、6、9天三个不同时间段支配树鼩眼内肌的副交感神经的兴奋状态;在实验的第6天分别处死模型组及正常对照组动物,取虹膜检测其cAMP(cyclic adenylate monophosphate,环一磷酸腺苷)、cGMP(cyclic guanylate monophosphate,环一磷酸鸟苷),并制作虹膜光镜、电镜片,进一步从第二信使、病理组织学方面观察支配树鼩眼内肌的副交感神经的兴奋状态。 结果:(1)树鼩瞳孔括约肌形态结构发育较为完善,较之大鼠、兔与人更为接近。(2)造模3天时,模型组和正常对照组树鼩瞳孔括约肌中MAChR的RT值及K_D值比较差异均无统计学意义(P>0.05);造模6天时,模型组树鼩瞳 成都?医药大学硕士学位论大孔括约肌中 MAChR发生增敏,KD值下降,即受体亲和力明显升高叩对.05人RT值未发生明显变化(P>0.05);造模9天时,模型组树勋瞳孔括约肌中MAChR发生上行性调节,RT值明显升高0m.05XKD值未发生明显变化叩川.05 )。 ()造模6天时,模型组树的瞳孔括约肌中CGMP含量明显增高(P<0.05),cAMP含量未发生明显变化(P>0,05)。(4)造模6天时,模型组树刚瞳孔括约肌胶原组织明显增多,HE染色平均光密度明显增高(P<0.01),平均黑度无明显变化(P>0.05);Masson染色平均光密度及平均黑度均无明显变化(P>0.05);”-8MA卜6mooth muscle action,。平滑肌肌动蛋白)染色平均光密度无明显变化(P>0.05),平均黑度明显增高 (<0.05)。 结论:结合前期实验研究结果,近距离久视所致支配树的眼内肌的副交感神经持续异常兴奋及交感、副交感双重神经支配平衡的失调,可能是肝劳发病的重要原因之一。
【Abstract】 Object: To discuss the reason of GanLao occurrence based on the experiment that analyzing parasympathetic nerve-receptor-second messenger and pathologic histology changes of iris sphincter muscle when accommodation continues.Methods:Tree Shrew was chosen as experiment object and was made staring continuously.After 3.6,9 days respectively,their iris were separate to quantitate the MAChR,cAMP,cGMP and to stydy pathologic histology changes.Results:(1)The iris sphincter muscle of Tree Shrew is more like human’s thanouse and rabbit. (2)After 3 days of staring,the MAChR in the iris sphincter muscle of Tree Shrew wasn’t change distinctly; After 6 days of staring,the MAChR was hypersensitivity; After 9 days of staring,the MAChR was up-regulated.(3) After 6 days of staring,the cGMP in the iris sphincter muscle of Tree Shrew increased, cAMP in the iris sphincter muscle of Tree Shrew wasn’t change distinctly;(4) After 6 days of staring,the collagen and the a -SMA in the iris sphincter muscle of Tree Shrew increased.Conclusion:With the results of the former study, the persistent erethism of parasympathetic and the disorder of the balance of sympathetic and sarasympathetic double innervation controlled iris sphincter muscle and ciliary muscle may be one of important reasons of GanLao occurrence.
