丝素蛋白粉的理化性质及其微胶囊的制备研究

【摘要】 丝素蛋白经酸法和碱法降解后脱盐是一个比较关键的步骤。它影响丝素粉的产率和成份。自制设备明显优于袋透析,使得氨基酸种类丰富且能够同时起到脱色的作用。经喷雾干燥的丝素粉呈球形颗粒,水溶性好。采用自制脱盐设备和喷雾干燥相配套制备丝素粉的工艺技术,不仅简便可行,而且将大大提高生产效率和经济效益。解决了制备丝素粉工艺过程中的“瓶颈”卡口,为规模化生产丝素蛋白粉提供了保障。 丝素胶体溶液具有良好的乳化特性和乳化稳定性,在pH3~pH10的范围内对其乳化性能和乳化稳定性影响不十分明显。丝素胶体溶液具有一定的起泡能力,且稳定性较好,在相同条件下,起泡能力不如酪蛋白酸钠,但稳定性较好。丝素乳化后在浓度较适宜的情况下具有包覆油相的网状结构。 丝素胶体溶液所具有的乳化特性为其应用于微胶囊的研究提供依据。包覆维生素E的丝素微胶囊具有良好的物理结构特征,不同的丝素浓度所需的芯材量不同,1%(质量浓度)的丝素溶液与芯材的比为1:0.6时比较合适。囊径在3μm~5μm之间,囊壁在500nm~1μm之间,囊壁经高温转为结晶结构特征时,更有利于芯材物质的缓释和渗透。

【Abstract】 In this dissertation, methods of degradation and desalination of silk fibroin were researched. The results showed that self-made equipments not only can increase efficiency of fibroin power producing and reclaiming, but also have the effect of discoloring. There is no obvious difference in amino acid content between fibroin power and rude fibroin. The grains achieved by spray drying are smaller than those achieved by freeze drying. So the method of using self-made equipments together with spray drying are simple and feasible.Fibroin protein solution, decomposed from degummed silk by calcium chloride, has finer emulsification ability and stability. Its these ability are not be obviously effected by pH value which is between 3 and 10.With fibroin solution concentration increasing, its emulsification ability , spume ability and stability are also increased. Solution system color is ivory-white like milk.Fibroin solution has perfect emulsification and stability, it also has the same important characteristic to the colloid. So it can be used to prepare for microcapsule. Fibroin microcapsules’wall have different structure characteristic with different fibroin solution. But these characteristics also effect on microcapsules release ability.
