扇贝多肽的透皮吸收及抗氧化机制研究

【摘要】 目的 制备新西兰大白兔的全皮、表皮和真皮的透皮吸收模型,采用该模型探讨以海洋生物工程技术自栉孔扇贝中提取出的扇贝多肽(PCF,相对分子质量800—1000Da)的透皮吸收及其抗氧化机制。方法 采用离体透皮试验装置,利用RP—HPLC法测定接收液中PCF浓度,通过公式计算PCF的单位面积累积透过量,进而通过回归方程求算出PCF的渗透系数,探究PCF在不同的时间、不同的浓度(分别为0.5%、1.0%、2.0%)、不同的pH值(pH分别为5.0、6.0、7.0)、有无透皮促进剂(透皮促进剂Azone的浓度分别为1%、2%、3%)及不同皮肤状态下的透皮吸收,并用化学发光法和ESR技术检测PCF对超氧阴离子(O2-)、羟自由基(OH·)及过氧化亚硝基(ONOO-)的清除作用以及其抑制亚油酸过氧化的作用。结果 结果表明:随着时间增加,PCF的单位面积累积透过量相应增加,2%浓度组的1h、2h、4h、8h、12h、24h的单位面积累积透过量分别为1.47μg、2.71μg、5.48μg 11.20μg、17.53μg、32.28μg;其透过皮肤的过程符合Fick’s扩散方程:随着PCF浓度增加,其24h单位面积累积透过量和渗透系数明显增加,0.5%、1.0%、2.0%三个浓度组的24h单位面积累积透过量分别为18.39μg、23.18μg、32.28μg;渗透系数分别为0.77μg.(cm2.h)-1、0.94μg.(cm2.h)-1和1.35μg.(cm2.h)-1;pH值为6.0时,24h单位面积累积透过量和渗透系数最大,5.0、6.0、7.0三个pH组的24h单位面积累积透过量分别为23.72μg、32.28μg、23.82μg,渗透系数分别为0.99μg.(cm2.h)-1、1.35μg.(cm2.h)-1、1.07μg.(cm2.h)-1;渗透促进剂Azone在2%时PCF的24h单位面积累积透过量和渗透系数最大,1%、2%、3%三个Azone浓度组的24h单位面积累积透过量分别为56.30μg、115.02μg、82.07μg,渗透系数分别为2.33μg.(cm2.h)-1、4.79μg.(cm2.h)-1、3.42μg.(cm2.h)-1;PCF透过角质层与全皮渗透差异较小,第三章中文摘要灰h单位面积累积透过量为32.96林g,渗透系数为1 .40林9.(c mZ.h).l;当去除角质层后,真皮层24h单位面积累积透过量为51.44卜g,渗透系数为2.11林9.(cm“.h)一,较完整皮肤增加560,0。PeF清除02·的Eeso一o.3omg·mL一l,抑制ONOO一的氧化活性的EC50=过氧化的抑制率为43%。结论020mgmL’清除oH·的Ees。一0.20 09·mL一‘,1.25mgmL一’的PcF对亚油酸脂质扇贝多肤在体外能透过全皮、表皮和真皮模型,并且受到时间、浓度、pH、透皮促进剂的影响,但透过皮肤模型的量很少,最高也只有总量的O,82%,其发挥抗氧化作用主要在皮肤表面和角质层;PCF能够有效清除自由基、抑制亚油酸的过氧化,是强的抗氧化剂。

【Abstract】 OBJECTIVE The polypeptide (PCF, Mr:800-l000) was isolated from Chlamys farreri using enzyme engineering technique. We studied the percutaneous absorption of PCF and the scavenge effects on free radicals in vitro. METHODS The concentrations of PCF filtered from the skin were detected with HPLC method on percutaneous absorption equipment in vitro. We first calculated the piled up percutaneous ratio of per area of PCF, then the permeating coefficient and the percutaneous absorption of PCF in different concentrations, pH values, with or without absorption promoter and different status of skin. Using chemiluminescence and electron spin resonance (ESR) techniques, we examined the scavenging properties of PCF on superoxide anions (O2 -), hydroxyl radical (OH-), peroxynitrite (ONOO-) and the inhibiting capacities of it on peroxidation of linoleic acid. RESULTS The results showed that the permeating coefficient of PCF increased obviously following the concentration increasing. At pH=6.0, with the absorption promoter Azone (2%), the permeating coefficient of PCF got to maximum respectively. PCF could scavenge oxygen free radicals including superoxide anions (O2 -) (EC5o=0.3mg/ml), hydroxyl radical (OH-) generated from the reaction systems (EC5o=0.2 g/ml) and effectively inhibit the oxidative activity of ONOO" (EC50=0.2mg/ml). The result of ESR indicates that PCF could inhibit the lipid peroxidation of linoleic acid notably. When the concentration of PCF was l.25mg.ml-1, the inhibitory ratio of PCF on the lipid peroxidation of linoleic acid was 43%. It is difficult to find another antioxidant which can scavenge so effectively both ONOO" andoxygen free radicals simultaneously. CONCLUTION PCF could hardly filtered and its percutaneous absorption was influenced by its concentrations, pH value, absorption promoter and the status of skin. PCF could scavenge free radicals and inhibit the peroxidation of linoleic acid in vitro. It may be a potential antioxidant from the marine products.
