利用SSR分子标记鉴定杂交稻种子纯度的研究

【摘要】 以18个在生产上大面积推广的水稻杂交种及其亲本为材料,分别利用SSR分子标记技术和田间种植表型观察进行纯度鉴定,通过比较两种鉴定方法的结果,探讨SSR分子标记鉴定的准确性和可行性。主要结果如下: 1.用20对SSR引物对18个杂交种的亲本材料进行SSR扩增,平均每个杂交种材料筛选出1-4对多态性较好的SSR引物。 2.用筛选出的引物对2001年春成都种植的20个试验材料进行SSR分析,并且将其分析结果与田间鉴定进行比较,结果发现两种纯度鉴定方法的结果在统计上不存在显著差异。 3.为了进一步比较SSR与由间鉴定方法的差异,判断SSR鉴定代替田间鉴定的可靠性和可行性,2001年增设重复并把田间实验扩大到了2个环境(海南和成都)。3种情况下的鉴定纯度的相关与回归分析结果为:SSR标记与海南试点结果的相关系数为0.785**,SSR标记与成都试点结果的相关系数为0.645**,成都与海南试点结果的相关系数为0.668**;分别用海南与成都试点鉴定结果作为依变量(Y1、y2),SSR鉴定结果为自变量(x)建立的一元回归方程为y1=32.70+66.4x和y2=31.80+67.4x,可对用SSR鉴定代替田间鉴定提供参考。 通过上述三个实验可以看出,两种鉴定方法(SSR分子鉴定与田间形态鉴定)在总体结果上不存在显著差异。随着市场经济的发展,传统的田间鉴定方法终将被SSR分子鉴定所取代,服务于农业生产,但有必要进一步优化鉴定体系。

【Abstract】 Eighteen rice hybrids and their parents were chosen to identify the hybrids’ purity with SSR markers and phenotype. The two methods of purity identification were compared in order to research the availability and accurateness of SSR marker identifying the rice seed’s purity .The results as follows:1. Eighteen rice hybrid parents’ DNA were amplified by twenty SSR markers, one or four SSR primers performed polymorphism between the double parents to average every hybrid.2. No significant difference was found between the average purity identified by SSR marker and the field experiment with 20 rice hybrids which were planted at Chendu in spring in 2001.3. In order to compare the difference between the two method strictly and estimate the availability and accurateness, the field experiment were be expanded two environment and three replication ,the SSR method also be expanded three replication. The results of correlation and regression analysis show that the correlation coefficient were 0.785**,0.645** and 0.668" respectively between the SSR marker and Hainan, SSR and Chendu, Hainan and Chendu.The average purity identified by experiment in Hainan and Chendu were chosed dependent variable(yi ,y2) and the average purity identified by SSR marker were chosed independent variable(x) , the regression equation were y1=32.70+66.4x and y2=31.80+67.4x.These results would be provided reference for the seed purity identification by SSR marker.In summary ,no difference founded in seed average purity identified by two methods of the SSR marker and phenotype. Field identification as the traditional method of seed purity identification may be substituted by SSR marker with the development of market economy, but some problem of SSR marker still be explored in practice.
