洋金花中治疗银屑病有效部位的化学成分研究

【摘要】 洋金花为茄科Solanaceae曼陀罗属植物白曼陀罗Datura metel L.的干燥花。其性温味辛、有毒,具有平喘止咳、镇痛解痉之功效;主要用于防治慢性气管炎、支气管哮喘,亦可治疗银屑病、有机磷中毒等病症,并用作外科手术的麻醉剂。近年来临床应用洋金花注射液治疗银屑病取得了显著的治疗效果。该方法具有治愈率高、复发率低等优点。 本文对洋金花治疗银屑病这一临床应用进行了有效部位的确定,并对有效部位的化学成分进行了深入系统的成分分离和结构鉴定研究。实验过程中采用732(001×7)型阳离子交换树脂和AB-8型大孔吸附树脂对50%乙醇洗脱组分进行了分离和精制,继而应用硅胶柱色谱、反相柱色谱、聚酰胺柱色谱等方法从中分离得到6种单体化合物,根据它们的理化性质和各种光谱(1H-NMR、13C-NMR及DEPT等)数据的解析,确定6种化合物的结构分别是山柰酚(Kaempferol)、7-O-α-L-鼠李吡喃糖基-山柰酚(7-O-α-L-rhamnopyranosyl-Kaempferol)、7-O-β-D-葡萄吡喃糖基-山柰酚(7-O-β-D-glucopyranosyl-Kaempferol)、3-O-β-D-葡萄吡喃糖基、7-O-α-L-鼠李吡喃糖基-山柰酚(3-O-β-D-glucopyranosyl、7-O-α-L-rhamnopyranosyl-Kaempferol)、3-O-α-L-鼠李吡喃糖基(1→6)-β-D-葡萄吡喃糖基、7-O-β-D-葡萄吡喃糖基-山柰酚[3-O-α-L-rhamnopyranosyl(1→6)-β-D-glucopyranosyl、7-O-β-D-glucopyranosyl-Kaempferol]、β-D-葡萄吡喃糖基(1→2)-β-D-葡萄吡喃糖基-苯乙醇[β-D-glucopyranosyl(1→2)-β-D-glucopyranosyl-Phenethylalcohol],其中化合物4、10和14均为首次从洋金花中获得。 本文研究结果不仅具有重要的理论价值,同时也为进一步将洋金花治疗银屑病的有效部位研制成二类新药提供坚实的基础,具有广泛的应用前景。
【Abstract】 Datura is dry flower of Datura metel L. It belongs to the family of Solanaceae. The drug is warm in nature and pungent in flavour. It has the effects of arresting cough and relieving asthma, relieving spasm and alleviating pain. In the clim’c,the drug is commonly used to treat chronic bronchitis bronchiad asthma and psoriasis.The injection of Datura has been used for the treatment of psoriasis these years and it showed an excellent curative effect. The method has advantages such as high cure rate, low relapse rate and so on.Clinical application of treatment psoriasis was identificated, and gives systemic studies, chemical constituents have been isolated and identified.By ion exchange resin 732(001 x 7) and AB-8 Macropore Adsorbing resin with silicagel column chromatography, ODS column chromatography and polyamide chromatography, six compounds (1-6) have been isolated from the 50% effective fraction of treatment psoriasis, using 1H-NMR, 13C-NMR, DEPT etc., we have indentificated chemical structures of six compounds:Kaempferol, 7-O-α -L-rhamnopyranosyl-Kaempferol, 7-O-→ -D-glucopyranosyl-Kaempferol, 3-O-α -D-glucopyranosyl, 7-O-α -L-rhamnopyranosyl-Kaempferol, 3-O-α-L-rhamnopyranosyl(l → 6)-β -D-glucopyranosyl,7-O-β -D-glucopyranosyl-Kaempferol, β -D-glucopyra nosyl(1→2)-β -D-glucopyranosyl-Phenethyl alcohol. The compounds 4,10 and 14 were isolated from Datura metel L. for the first time.The result of this article not only has important value in theory, but also provides foundation for making effective fraction of Datura metel L. into a new drug.
