健骨灵对实验性骨质疏松大鼠雌激素受体mRNA及Ⅰ型胶原mRNA表达的影响

【摘要】 骨质疏松症(Osteoporosis,OP)是一种以骨量减少、骨组织显微结构退化为特征,以致骨的脆性增高而骨折危险性增加的一种全身性骨病。本病多发于中老年人和绝经后妇女,可发生腰椎压缩性骨折,或在不大的外力下发生桡骨远端、股骨近端和肱骨上端骨折,极大的影响了患者的生存质量。国内外对本病的研究众多,进展很快,从单纯的补钙到基因治疗有了很大的突破。目前,西医研究表明激素替代疗法是治疗骨质疏松症较为有效的方法。但激素替代疗法在给患者带来危险因素方面存在很大争议。而中医药因其毒副作用小的特点,越来越引起人们的关注。本实验从分子水平出发,从而探讨中药健骨灵治疗骨质疏松症的机制。 为了探讨健骨灵治疗骨质疏松症的分子作用机制,本研究以成年雌性卵巢切除大鼠作为骨质疏松症实验模型,通过HE染色研究中药健骨灵对骨组织的成骨细胞数目的影响;通过原位杂交技术,在mRNA分子水平研究,雌激素受体mRNA及Ⅰ型胶原mRNA分布及表达。结果显示,中药健骨灵可使成骨细胞数目增加(p<0.01);原位杂交显示雌激素受体mRNA及Ⅰ型胶原mRNA主要分布于成骨细胞胞浆中,并且运用健骨灵治疗后雌激素受体mRNA及Ⅰ型胶原mRNA表达水平提高(p<0.01)。根据本实验结果提示,中药健骨灵可以起到雌激素样作用,通过对雌激素受体mRNA及Ⅰ型胶原mRNA调控,从而促进骨形成。

【Abstract】 Osteoporosis is a systemic skeletal disease characterized by low bone mass and microarchitectural deterioration of bone tissues, with a consequent increase in the bone fragility and the susceptibility to the fracture. The wrinklies, the edlies and postmenopausal women suffered from osteoporosis that may lead to the compressive fractures of lumbar vertebrae or the distal radial fractures ,the proximal femoral fractures and the superior humeral fractures ,which affect the existent qualities of the patients to a great extent. The fractures occurred lumbar vertebrae. And it was legion and fast that the studies on osteoporosis which breakthrough were prodigious from dose of calcium to the remedy of gene. Currently, studies of western medicine indicated that the HRT was the best way to OP. There was a controversial aspect of risk of cancer to patients yet. Because of its less poisonous side effect, the traditional Chinese medicine more and more was paid attention to. This experiment investigated the mechanism of JIANGULING on osteoporosis in the molecule level.To investigate the molecular links of the treatment with JIANGULING on postmenopausal osteoporosis in vivo, adult ovariectomy rats were used in this study as the model of postmenopausal osteoporosis. The numbers of osteoblast in the bone tissues were analyzed using HE staining; Estrogen mRNA and type I collagen mRNA expressions in bone tissues of ovariectomy (OVX), ovariectomy with diethylstilbestrol, ovariectomy with JIANGULING and ovariectomy with GUSHUKANG rats were analyzed at mRNA level using Tissue In Situ Hybridization (TISH). The results showed that the expressions of both ERmRNA and COLLA I mRNA were higher than those of VOX in the bone tissues(p<0.01). The numbers of osteoblasts of OVX-JIANGULING were more than the those of OVX (p<0.01).The tissues in TISH showed that ERmRNA and COLLA I mRNA were mainly expressed in osteoblasts. These datum indicated JIANGULING played an important role called " estrogen-like " , which prompted bone formation by regulating ERmRNA and COLLA I mRNA.
