淋巴细胞对子宫内膜蜕膜化及胚胎着床行为的影响

【摘要】 目的:探讨淋巴细胞对子宫内膜蜕膜化及其胚胎着床行为的影响。 方法:1.分离育龄期妇女增殖晚期及分泌期子宫内膜间质细胞,加入雌二醇(E2)、孕酮(P)使其蜕膜化,分别加入人黄体期及早孕期外周血淋巴细胞共培养,观察蜕膜化的间质细胞生长情况并测定培养液中PRL含量。2.培养8天后,加入小鼠第四天胚胎继续培养,观察胚胎的孵化、黏附、扩展生长情况及间质细胞生长情况。 结果:1.加入早孕期淋巴细胞的子宫内膜间质细胞蜕膜化程度明显优于黄体期淋巴细胞添加组和对照组。2.早孕期淋巴细胞添加组分泌PRL的量与对照组相比有显著性差异(p<0.05),而黄体期淋巴细胞添加组虽然分泌PRL的量高于对照组,但无统计学差异。3.黄体期及早孕期淋巴细胞均能明显促进胚胎在子宫内膜间质细胞上的孵化、黏附、扩展性生长(p<0.05),尤其以促进孵化和黏附为明显(p<0.01),早孕期淋巴细胞添加组胚胎扩展生长率显著高于黄体期淋巴细胞添加组(p<0.05)。 结论:1.早孕期淋巴细胞对子宫内膜间质细胞蜕膜化有明显促进作用,特别是特异性促进PRL的分泌。2.早孕期淋巴细胞明显促进胚胎在子宫内膜间质细胞共培养体系中的扩展性生长。3.早孕期淋巴细胞有可能通过促进E2、P激素调节下发生蜕膜化的子宫内膜分泌PRL而促进胚胎着床。

【Abstract】 Objictive: to investigate the roles of lymphocytes in regulation of endometrial stroma cells decidualization and embryo implantation.Methods:1.Stroma cells derived from human endometrium during the late proliferate phase or secretive phase were treated with estradioK progesterone to induce decidualization. Peripheral blood lymphocytes(PBL) isolated from women in the luteal phase and early pregnancy were added to to the stroma cells. The stroma cells morphologic changes and prolactin (PRL)production in the media were examined and compared.2. The 4-day-mouse embryo were added to the stroma cells after cultured 8 days. The hatching, adhereing, outspreading of mouse embryo and the development of stroma cells were observed under the microscope.Results: 1. PBL derived from early pregnancy women significantly increased the degree of the stroma cells decidualization compared with PBL derived from luteal phase and the control group.2. PRL secretion of co-cultured with PBL isolated from early pregnancy women was significantly higher than the control group(p<0.05). Wheras the significantly augmentation of PRL prouduction was not observed in co-cultured with PBL isolated from women in luteal phase. 3.PBL isolated from luteal phase women and early pregnancy women significantly promoted the embryo hatching, adhering, outspreading on the endometrial stroma cells(p<0.05),esperialy the hatching and adhering(p<0.01). PBL isolated from early pregnancy women significantly promoted embryo outspreading compared with PBL isolated from luteal phase women(p<0.05).conclusion:1.The lymphocytes derived from early pregnancy can contribute to the stroma cells decidualization and particularly improve PRL secretion.2. Lymphocyte isloted from early pregnancy can promote the embryo outspreading in co-cultured with endometrial stroma cells. .3. The lymphocytes derived from early pregnancy may promote the embryo implantation by improving PRL secretion based on stroma cellsdecidualization regulated by E, P.
