芦荟组培快繁技术及其遗传资源的RAPD分析

【摘要】 芦荟系百合科芦荟属多年生肉质常绿草本植物,原产非洲东南部及地中海沿岸一带,我国的南、北方均有种植,是一种很有开发前途的经济植物资源。尤其美国库拉索芦荟除药用外,在观赏、食品保健、美容化妆方面均有重要开发应用价值。本研究开展了芦荟组培快繁技术及其遗传资源的RAPD分析。 一、芦荟遗传资源的RAPD分析 本实验提取了纯度较高的芦荟基因组DNA,电泳结果见到一条比较亮的条带。对RAPD反应体系进行了优化,通过对模板DNA浓度、dNTPs浓度、MgCl2浓度、引物浓度、Taq聚合酶浓度逐个进行梯度实验,获得适合芦荟材料的RAPD反应参数。 本研究采用随机扩增多态性(RAPD)标记,探讨了13芦荟品种的遗传背景和多态性。对206个随机引物中对13个芦荟品种的DNA样品进行筛选结果表明,共获得了16个有稳定多态且带型清晰扩增带的引物,可用于芦荟的遗传变异多态性鉴定。35个引物共扩增出173条谱带,其中78条谱带具有多态性,每个引物可扩增出1~9条多态性谱带,平均4.94条。 二、芦荟组织培养快速繁殖 本实验以芦荟叶片和分蘖原基诱导不定芽、根的方法,建立了较完善的芦荟组织培养快速繁殖实验体系。通过对比较了2种培养基(MS培养基、N6培养基、各种浓度梯度的生长素NAA和分裂素6-BA,3个温度(20、25、30±2℃不同培养温度、3种光照条对芦荟不定芽快繁系数的影响,确立了芦荟组织培养快速繁殖的参数。并在实际应用中发挥了较大的作用,显示了良好的前景。
【Abstract】 Aloe vare L, growing mainly in the dry regions of Africa, Asia, Europe and America, is a member of the Lily family, it is very cactus-like in its characteristics. In the present study, vegetative propogation and analysis of RAPD of Aloe vare L were carried out. The primary results could be summarized as follows:1. The genome DNA from 13 families of Aloe were succesfuly extracted by using the improved CTAB method. And the analysis of genetic diversity of these families in Aloe vera L. were carried out by RAPD molecular markers, which may help to provide the theoretical base for breeding. 16 primers were screened from 206 random oligonucleotide primers and 39 polymorphic loci were obtained from PCR of 17 primers. 35 of the primers could amplify polymorphic bands. Out of 123 amplified bands, 78 bands are polymorphic. Every primer could amplify 1 to 9 polymorphic bands, with an average of 4.94 bands.2. Significant differences were observed among the different media, auxins, culture temperature, culture light. The regenerated plantlet percentage was significantly higher under medium MS, under 25 ℃ and 12h light/12h dark than that under medium N6, 20℃ and 30℃and 24h dark. No difference was observed between under 20 ℃ and 30℃, as well as 12h light/12h dark and 24h light. On the basis of the results, a system for vegetative propogation of Aloe vera L was established and applicated in the farm practice.
