金银花绿原酸的提取分离及其功能产品开发

Separation of Chlorogenic Acid from Honeysuckle and Its Development of Functional Product

【摘要】 本论文采用酸性乙醇回流法和大孔吸附树脂分离技术相结合的方法提取分离金银花绿原酸,对其最佳工艺参数进行了研讨,然后以绿原酸为主研制出具有抗菌抗氧化作用的气雾剂复方。 通过正交试验得到回流提取绿原酸的工艺参数:提取溶剂为70%乙醇、提取溶液的pH值为4、回流时间为2h,在此条件下,绿原酸得率为5.32%,产物中绿原酸含量为36.13%。活性炭脱色的工艺参数为:活性炭添加量为1.25g/100mL绿原酸浓缩液、脱色温度为60℃、脱色时间为30 min、绿原酸浓缩液pH值为3,在此条件下,活性炭脱色率达到85%以上。通过对比8种大孔吸附树脂对绿原酸的吸附效果,选用对绿原酸吸附解吸性能均较好的树脂对绿原酸吸附,吸附参数如下:吸附液绿原酸的浓度为0.204mg/mL、吸附液pH值为4、流速为3mL/min,解吸参数如下:解吸剂为30%乙醇、解吸剂流速为1.5mL/min、解吸剂使用量为4倍树脂体积(V/V),在此条件下,绿原酸得率为5.06%,产物中绿原酸含量为78.26%。提取中药材连翘、甘草、桔梗、麦冬中的活性成分连翘酯甙、甘草酸、桔梗皂甙、麦冬多糖与绿原酸组合,研制成复方气雾剂。经体外功效试验绿原酸对金黄色葡萄球菌有较强的抑制作用,对DPPH自由基有较强的清除作用;气雾剂复方对肺炎链球菌、金黄色葡萄球菌、甲型链球菌等肺部感染病原菌有较强的抑制作用。

【Abstract】 In this paper, the reflux method of ethanol with acidity and the technique of separation with macro resin were used to extract and separate chlorogenic acid (CHA)from Flos Lonicerae. The optimal condition of reflux extraction and the parameters of adsorption were thoroughly studied.By orthogonal design, the optimal condition of extraction is: the ethanol concentration is 70%, the ethanol acidity (in terms of pH) is 4, the reflux time is 2 h. In this condition, the final yield of CHA is 5.32%; the content of CHA is 36.13% in the production. The condition of adsorption with active carbon is: the addition amount of active carbon is 1.25 g per 100 mL solution, the adsorption temperature is 60 癈, the adsorption time is 30 min, the acidity of adsorption solution (in terms of pH) is 3. In this condition, the color of adsorption solution was decreased 85% above. By comparison of eight micro adsorption, A resin with high adsorption rate and eluent rate was used to adsorb chlorogenic acid from extraction solution. The adsorption parameters are: the content of chlorogenic acid is 0.204 mg per mL adsorption solution, the acidity of adsorption solution (in terms of pH) is 4, the adsorption velocity of flow is 3 mL per min, the eluent is 30% ethanol, the eluting speed is 1.5 mL per min and the eluents volume is 4 times of resin volume. In the optimal adsorption condition, the final yield of CHA is 5.06%. the content of chlorogenic acid is 78.26% in dry productin. Several Chinese herbal medicine were selected to combine aerosol compound, such as Honeysuckle, Forsythia suspense Vahl, Glycyrrhiza uralensis, Platycodon grandi florum and Ophiopogon japonicus. The anti-bacterial and anti-oxidation tests were done to testify the effect of aerosols compound. By in vitro functional experiment, CHA has obvious effect of grownth inhibition on staphylococcus aureus and a cleaning DPPH free radical. Aerosols compound has significant in vitro effect of grownth inhibition on streptococcus pneumonriae^ staphylococcus aureus, a -hemdytic streptoccus etc.
