黄瓜自交系及其F1代的RAPD分析

【摘要】 本研究结合田间实验,利用RAPD标记从分子水平上探测了黄瓜(Cucumis sativus)亲本自交系与其杂种F1代的遗传差异。用30个引物对11个黄瓜基因型进行了扩增,共检测出234个位点,其中多态性位点167个,占总位点的71.37%。,每条引物可扩增出2—17条带,平均每条引物产生7.25条带。根据RAPD遗传距离矩阵,按UPGMA方法进行了聚类分析,结果表明:杂种F1代偏向母本自交系遗传,由亲本自交系间遗传距离可见,黄瓜亲本自交系间的遗传距离在0.123—0.164之间,说明黄瓜是一种遗传变异较小的蔬菜。做为父本,津研七号自交系与宁阳大刺自交系相比,子代更多继承了津研七号自交系的遗传特性。以上结果与田间性状表现及实际情况基本相符。RAPD标记用于黄瓜遗传差异的研究切实可行,其结果可用于育种过程中的亲本选配和分子标记辅助选择。
【Abstract】 This study detected genetic differences between cucumber ( Cucumis sativus L.) inbred lines and its’ hybrid progenies F1 from molecular level by means of RAPD marker combing field experiment. A total of 30 primers were employed for PCR amplification of 11 genotype of cucumbers, from which 167 polymorphic loci were stored, about 71.37% of total loci. The polymorphic products amplified by each primer ranged from 2-7, with an average of 7.25. According to RAPD genetic distance matrix, cluster analysis was carried out by UPGMA method. The result showed that the hybrid progenies F1were inclined to female inbred lines in heredity. Genetic distance among parents inbred lines ranges form 0.123 to 0.164. The result showed that the genetic differences of cucumber were small. As male, F1 inherits more genetic characteristics of Jinyan7’ inbred lines ’ than Ningyangdaci inbred lines . The above results are roughly concordant with field experimental result and actual situation. It is feasible for RAPD to use for studying genetic diversities of cucumber. The research result can act as theoretical basis for parental selection and molecular marker-assisted selection in breeding procedures.
