三氧化二砷对肝癌细胞株BEL-7402和SMMC-7721的作用及机制探讨

【摘要】 目的:观察不同浓度三氧化二砷作用不同时间对肝癌细胞株BEL-7402和SMMC-7721的影响并探讨其作用机理。 方法:应用不同浓度三氧化二砷作用于肝癌细胞系BEL-7402和SMMC-7721不同时间后,台盼蓝拒染法计数活细胞;Gimsa染色法观察细胞形态;流式细胞仪检测细胞DNA含量;琼脂糖凝胶电泳观察凋亡梯状条带;端粒酶检测试剂盒检测端粒酶活性的变化;药物干预BEL-7402和SMMC-7721细胞内谷胱甘肽含量,比较两种细胞对三氧化二砷诱导凋亡敏感性的差异。 结果:0.25~2.00μmol/L三氧化二砷可显著抑制肝癌细胞系BEL-7402的生长,观察到凋亡的形态学改变,流式细胞仪检测亚G1期细胞明显增多,端粒酶活性下降,抑制作用与时间、剂量呈依赖性关系。0.25μmol/L三氧化二砷作用96h对SMMC-7721细胞生长和端粒酶活性没有影响。0.50~2.00μmol/L三氧化二砷可抑制SMMC-7721细胞端粒酶活性,与时间、剂量呈依赖性关系;1.00~2.00μmol/L可显著抑制SMMC-7721细胞生长,促进细胞凋亡。BEL-7402细胞内谷胱甘肽含量比SMMC-7721细胞低,三氧化二砷诱导BEL-7402细胞凋亡的百分率较后者变化明显。提高细胞内谷胱甘肽的含量可降低两种肝癌细胞对三氧化二砷诱导凋亡的敏感性。 结论:三氧化二砷可以抑制肝癌细胞系BEL-7402和SMMC-7721的生长,其机制可能是诱导凋亡和降低细胞端粒酶活性。干预细胞内谷胱甘肽含量可以改变BEL-7402和SMMC-7721细胞对三氧化二砷的敏感性。
【Abstract】 Methods: Hepatoma cell lines BEL-7402 and SMMC-7721 were treated with different concentrations of As2O3 in combination with various time; viable cells were determined by trypan blue exclusion; apoptotic cells with Gimsa staining were observed under microscope and agar electrophoresis was used to assay DNA from apoptotic cells, and DNA contents of apoptotic cells were determined by flowcytometry; telomerase activity of cells treated by As2O3 was assayed by Telomerase PCR ELISAPLUS Kit and GSH contents of them were detected so as to compare the sensitivity between them to As2O3.Results: From 0.25 to 2.00μmol/L, As2O3 can inhibit growth of BEL-7402 cells significantly with concomitant of apoptosis, increasing sub-G1 phase cells, and decrease its telomerase activity, which showed dose-dependent and time-dependent effects. There is no any effect of As2O3 on SMMC-7721 cells at level of 0.25μmol/L. However, there exist effect of As2O3 mentioned above at level of 0.50 to 2.00μmol/L. The experimental result also showed that GSH involved in sensitivity of BEL-7402 and SMMC-7721 cells to As2O3, the higher level of GSH content in cells, the lower sensitivity of cells to As2O3.Conclusion: As2O3 can inhibit hepatoma cell lines BEL-7402 and SMMC-7721 cells growth, the mechanisms perhaps are to induce apoptosis and to decrease their telomerase activities. The GSH content can effect the sensitivity of them to As2O3.
