鲁北白山羊早期卵泡发育及子宫输卵管的组织学研究

【摘要】 山羊的单胎产仔性能介于单胎动物和多胎动物之间,其生殖特性的研究有助于从理论上对动物生殖生理的深一步认识,也为提高动物的繁殖率奠定理论基础。不论是哺乳动物的克隆技术的研究和应用还是胚胎体外生产,对具有发育潜力的和受精能力的高质量的卵母细胞需求量日益增加。进一步开发和利用家畜原始卵泡的大量而丰富的资源,已是当前畜牧繁殖生物学的热点之一。为了研究家畜的原始卵泡资源,探索多胎性状与卵泡发育的关系,同时为进一步开发和利用家畜的原始卵泡资源提供可靠的数据资料。本文以鲁北白山羊为主要研究材料、以东北细毛羊为对照材料,对3月龄山羊卵巢的卵泡发育及山羊和绵羊子宫输卵管组织学结构进行了研究。实验表明:3月龄山羊卵巢最大切面的皮质部分卵泡密度为17.45±7.36个/mm~2,3月龄原始、初级卵泡是一个椭圆形的外部结构,长、短轴直径约为19.60±4.00、15.05±3.18μm和65.06±15.60、52.12±14.13μm。山羊卵泡的透明带是在中间卵泡的Ⅰ阶段开始形成的,但此时并不完整。3月龄山羊卵巢中的原始卵泡、初级卵泡出现多卵母细胞(MOFs)以及双核的有腔卵泡现象。山羊和绵羊的输卵管上皮纤毛具有典型的9+2微管结构。实验揭示了山羊原始卵泡的形成过程以及3月龄山羊卵巢中原始卵泡窝分裂的动态过程研究。同时对绵羊卵巢原始卵泡的卵泡细胞的来源、募集、分裂以及迁移动态进行了研究。

【Abstract】 The goat in lambing trait is between single-fetus and multi-fetus animals.The study on its reproductive trait is useful for us to further understand animal reproductive physiology theoretically and lay theoretical base for improving the efficiency of animal reproductive rate.The oocytes with high fertilizing capacity and developing competence are required steadly in the research on mammalian cloning as well as in vitro embryonic production. Now, it is a hotspot in the biology of animal reproduction to exploite a large number of primordial follicles of livestock in ovary. The objects of experiment aimed at research on the primordial follicular resource of livestock, studing the relations between multi-fetus trait and follicular development and providing useful data for further exploiting the primordial follicular resource of livestock. Lubei white goats were used with the fuzz sheep of Northeast as contrast. The ovaries of three-month old goat and uterus and oviduct of sheep were studied in histological structure.The results show as followed: the density of cortes primordial follicles is 17.45?.36/mm in the largest cross section; The primordial and primary follicles take the elliptic shape in cross section. The diameter of long and short axis for both follicles are 19.6 4.00, 15.05 3.18 and 65.06 15.60, 52.12 14.13um respectively; The ZP of goat ovaries appear in the first phase of intermediary follicles. The primordial follicles with multi-oocyte and twin-oocyte antral follicles have been found in three-month old goat. The cilia epithelium of goat and sheep oviduct has a typical 9+2 micro-tubal structure. It was described that the forming course of goat primordial follicle, the division dynamic process of primordial follicles nest, the origin ^ collect^ division and migration of the follicle cell of primordial follicle in sheep ovaries,
