基于ALA的PDT体外灭杀白血病肿瘤细胞新方法研究

【摘要】 光动力疗法(Photodynamic Therapy,缩写PDT)是利用细胞和生物组织内部发生的光化学相互作用/光动力作用进行治疗的方法。本论文第一部分介绍了国内外光动力疗法的研究状况,并阐述了本课题的研究意义和主要研究内容。论文的第二部分对白血病的基本病理做了简要的阐述,并综述了目前治疗白血病的常规方法。第三部分对光动力疗法的原理、PDT中的光敏剂及光源做了详细的论述。第四部分介绍了我们创建的ALA-PDT仪器系统。第五部分用我们的ALA-PDT仪器系统对白血病肿瘤细胞HL60进行了实验研究,并获得了一系列优化实验参数。第六部分对全文进行了总结。 本论文的主要研究工作有: 1.本文对白血病的基本病理做了综述并纵观了目前白血病的一些常规治疗方法。 2.本文对ALA-PDT的作用机理做了深入性的探讨,阐述了PDT中使用的光剂量、光波长及光动力疗法的毒副作用。 3.完成了ALA-PDT光反应室的设计及整个光动力疗法实验系统的组建。该系统使用普通光源进行辐照,能在400~650nm间进行多光波段的选择,输出功率在1~8mW间可调。 4.应用我们创建的PDT实验系统对白血病肿瘤细胞HL60进行了实验研究,取得了一系列ALA-PDT的优化参数,如:ALA的浓度、ALA的培养时间、辐照光剂量、光功率、辐照光波长及培养细胞的FCS浓度等。

【Abstract】 Photodynamic therapy, abbreviated to PDT, is a therapy which uses the photodynamic or actinic chemistry interrelation occurred in the cells. We introduce the research status about the PDT which takes place domesticly and overseas, and expatiate on the significance of our research and the content of our research at the first part of the dissertation. In the second part, the basic pathology of leucocythemia has been expatiated simply, and the conventionally therapies that have been currently applied in the treatment of leucocythemia are summarized. Followed, in the third part, we detailedly dissertate the principle of PDT > the photosensitizer and the lamp-house used in the course of PDT. In the forth part, it is introduced the experiment system of ALA-PDT which we established. In the fifth part, by use of the experiment system of ALA-PDT, we research the leukaemia cells - HL60 and achieve a serial of optimizational experiment parameters. In the last part, the whole dissertation is summed up.The major achievements made in this dissertation are as follows: 1 . The basic pathology of leucocythemia has been summarized and the conventionally therapy of leucocythemia was fully introduced.2. We probe deeply into the action mechanism of ALA-PDT and set forth on light-dose, light- wave used in the course of PDT and the noxious side-effect followed in the treatment of ALA-PDT.3. The author designed the light-reaction room of ALA-PDT and set up the whole PDT experiment system, which uses a common lamp-house and can be adjusted to choose multiwave bands from 400nm to 650nm and be output kinds of power below 8mW.4. By use of the experiment system of ALA-PDT, we research the leukaemia cells - HL60 and achieve a serial of optimizationalabstractexperiment parameters, such as: the density of ALA, the culture time of ALA , the shined-light dose , the shined-light power , the shined-light wavelength and the density of PCS in the course of cell culture.
