【摘要】 目的 为了进一步探讨针刺治疗中风病的机理及其治疗时机。方法 本实验应用栓线法栓塞大鼠大脑中动脉(MCAO)造成局灶性脑缺血模型，观察了急性期电针和恢复期电针对局灶性脑缺血大鼠神经体征的促恢复作用，并检测血液中一氧化氮(NO)、内皮素(ET)、超氧化物歧化酶(SOD)、丙二醛(MDA)的变化，以及电针对局灶性脑缺血大鼠神经元凋亡相关基因bcl-2、bax蛋白表达的影响。结果结果表明电针可明显促进神经体征的恢复，急性期电针促恢复作用比慢性期电针明显；电针可以明显降低脑缺血后升高的NO水平，减少NO的神经元毒性作用，逆转脑缺血后血浆中ET升高的趋势，从而改善脑血流量，防止脑缺血后神经元的进一步损伤；电针可以显著提高局灶性脑缺血大鼠血清SOD活性，降低血清MDA含量，抑制自由基的产生从而延缓神经细胞的缺血性坏死；电针能够诱导Bcl-2和抑制Bax的表达，从而抑制神经细胞的凋亡。结论 该研究从分子生物学、细胞学、组织学等多层次揭示了急性期及慢性期电针对MCAO大鼠的影响，显示急性期针刺组的治疗效果好于慢性期针刺组，提示临床针刺治疗缺血性中风应该从急性期开始。更多还原

【Abstract】 Objective To investigate the mechanism of acupuncture on cerebral ischemic vascular diseases and appropriate time for acupuncture.Methods The focal cerebral ischemia rats model are made by middle cerebral artery occlusion(MCAO) with nylon thread embolus .We observe the improgress of nerve signs in the acute and chronic ischemic stage after electroacupuncture, detect the change of nitric oxide(NO),endothelin(ET),super-oxide dismutase(SOD),MDA of blood.We also detect the expressions of bcl-2 and bax which are neuronal apoptosis related genes. Results The results indicate that electroacupuncture can accelerate the disappearance of pathological nerve sign. The recovery at the group of acupuncture in acute stage of cerebral ischemia rats is better than that at the group of acupuncture in chronic stage . Electroacupuncture can decline the amount of NO, and decrease the poisonous effects on focal cerebral ischemia rats. Electroacupuncture also can reverse the rising trend of ET in the blood, thus improve the flow of blood in brain and protect further damage of the neurons. Electroacupuncture can improve serum SOD activation and reduce MDA content notably of focal cerebral ischemia rats,thus suppress production of free radical and delay neuronal cell death .Electroacupuncture can lead the expression of Bcl-2 protein and suppress Bax protein , then suppress the apoptosis of neurons.Conclusion The experiments have announced that electroacupuncture affected MCAO rats in acute and chronic stage from molecularbiology,cytology,histology and other levels.The results shows that the effects of acupuncture in acute stage is better than that in chronic stage.lt suggests that clinical acupuncture for cerebral ischemic stroke should begin from acute stage.更多还原
