一种新型固相放免材料的研究

Study on a Kind of New Solid-phase Material for Radioimmunoassay

【摘要】 利用Hunter试剂的原理,将N-羟基琥珀酰亚胺(NHS)酯功能基团引入高分子载体上,合成了几种烯酸与苯乙烯聚合物的NHS酯。研究了合成烯酸NHS酯及聚合物的NHS酯的合成条件,测试了烯酸NHS酯及聚合物NHS酯的特性。 将聚合物的NHS酯涂于聚苯乙烯试管或小球表面获得能与蛋白共价结合的功能材料。研究了各种功能材料与蛋白结合的各种条件:反应温度,反应时间,介质pH值,蛋白的种类对结合的影响。实验证实,这类聚合物NHS酯可在温和条件下与蛋白反应,使蛋白以共价键结合方式固定在聚合物上,这种功能材料对蛋白的结合量大于1.5μ/mm~2,对蛋白的一次吸附率大于80%,能满足固相放免分析的要求,将为固相放免分析提供一种新材料。 本文还利用自制的固相碘标记活化珠进行了碘标记蛋白质的方法和效果的研究。考察了各种标记条件:活化珠用量及粒径、pH值、温度、标记时间对标记率的影响。结果显示,用此活化珠,碘标记条件温和,标记效率高,标记物稳定且对标记蛋白质的损伤小,适用范围大,因此具有良好的应用前景。

【Abstract】 Based on the principle of Hunter reagant,N-Hydroxysuccinimide(NHS) functional group was introduced into polymer carrier to synthesize a couple of olefine acids’ and styrene’ copolymer-NHS-esters.’ Conditions to prepare the NHS-esters of all olefine acids’ and polymer-NHS esters were studied. And these products’ basic physical and chemical properties were also tested.Self-prepared polymer-NHS-esters were coated on PS test tubes and small PS balls and got a kind of function material which can connect with proteins through covalent bounds. Various conditions of this material to bind proteins were studied: reaction temperature, time, media , protein types and so on . The test results showed that the amount of this kind of function material to conjugate proteins were larger than 1.5[ig/mm2and its one time adsorption rate was larger than 80% and can satisfy the requirements for solid-phase radioimmunoassay. Test results showed that this polymer-NHS-ester can connect with proteins through covalent bounds under mild conditions This technical method will supply a type of new material for solid-phase radioimmunoassay(RIA).The method and effect of using self-prepared solid-phase iodine-activated beads to iodine-label proteins were studied.The effects of various iodine-labeling conditions such as the using dose and size of activated beads ,pH value,temperature,labeling timeperiods ,the volume,and the dose of protein on labeling rate weretested.The results suggested that iodine-labeling can be performed under mild conditions and which has a wide range of applications and its labeling effectiveness is high and the substances labeled were quite stable and it has very little damage to the bio-activity of the labeled proteins.So it has a bright application prospect.
