导数光声光谱技术及其在生物医学上的应用

【摘要】 本论文第一部分概述了光声光谱技术的发展,尤其是光声光谱技术在生物医学上的应用。第二部分论述用光声技术测量人鼻咽癌的光谱吸收特性。第三部分提出导数光声光谱技术,将光声光谱技术和导数光谱结合起来,使导数光声光谱技术同时具有光声光谱技术的可适用于强散射,非透明的样品测量和导数光谱可分辨重叠光谱,分辨肩峰的优点,又克服各自的缺点。 本论文的主要工作有以下几个方面: 1.用自制的高灵敏度的传感器组成的测试系统测量了光电探测器的相对光谱响应。该实验表明我们的测试系统是线性系统,所测光谱数据可靠。 2.对常用的双光束归一化光声光谱技术进行改进,并进行实验研究。改进后的归一化方法消除了分光镜的影响,每次测量的数据具有可比性,实现了光谱数据的数字化,为导数光声光谱技术的实现准备条件。 3.用改进的归一化光声光谱技术测量了三株人鼻咽癌细胞(SLINE-1,CNE-1,CNE-2)的归一化吸收光谱系数以及这三株细胞在420nm处的吸收光谱系数对X光放射剂量的依赖关系,并用胎盘兰对细胞染色,用以反映死细胞的光声信号。 4.提出导数光声光谱技术,并用软件实现。并用导数光声光谱技术测量了各种人血的零阶,一阶,二阶导数,实验表明:导数光声光技术比普通光声吸收光谱技术具有更高的检测灵敏度;许多人的血液的光声谱在637nm和664nm附近有吸收峰。
【Abstract】 Emphasizing the application of the photoacoustic spectroscopy (PAS) technique in biology and medical, the development of the PAS technique is summarized at the first part of the dissertation. In the second part, that the spectrum absorption characteristic of opharyngeal carcinoma measuredPAS technique is discussed. In the last part, the differential PAStechnique, which combines the PAS technique with the differential spectrum, overcomes the defect from the two techniques, making it has the both advantage of the PAS technique, which is suitable for the measurement of the heavy diffusion > opacity sample, and the advantage of the differential spectrum which is capable of differentiating the overlap spectrum and shoulder peak, is present. The achievement made in this dissertation are as follows:1. The relative spectroscopy responsibility of the photoelectric detector was measured using the measurement system made up from the highly sensitive photoacoustic sensor made by ourselves. The experiment shows that our test system is a line system, the data we measured is reliable.2. The double beam normalizing PAS technique is improved and studied through the experiment. After improving, the effect of the beam splitter is removed, all the data measured can be compared together. The digital spectrum data is convenient for the implementation of the differential spectroscopy.3. Using the improved PAS technique, the relative absorption spectroscopy of three ppharyngeal carcinomas cell (SUNE-l,CNE-l,CNE-2), relative absorption coefficient at the wavelength of 420nm of the three cells exposure to different X light quantity, are measured. The photoacoustic signal from the died cells is studied through dying the cells.4. The differential PAS is present and accomplished. Using the new technique, the zero, the first and the second differential PAS of some persons’ blood was measured, the result shows: the new technique is more sensitive than the common PAS technique. The PAS of many persons’ blood has absorption peak around the wavelength of 637nm and 664nm.
