白芥子抑制前列腺增生的药学研究

【摘要】 本文综述了良性前列腺增生(BPH)的发病机理、药物治疗概况以及白芥子(seedsof Brassica alba)的药学研究概况,报道了白芥子各成分的提取、分离、含量测定、白芥子抑制前列腺增生的有效部位、有效物质的筛选及作用机理的研究结果。 建立了白芥子各类物质的最佳提取条件。 从白芥子提取物中分离得白芥子苷(sinalbin)、白芥子碱(sinapine)、β-谷甾醇(β-sitosterol)。 采用高效液相色谱法(HPLC)测定白芥子β-谷甾醇含量,采用气相色谱法(GC)、气质联用技术(GC-MS)分析测定白芥子脂肪酸成分与含量,采用氨基酸自动分析仪测定白芥子中氨基酸的成分与含量,采用硫酸钡沉淀法测定白芥子苷含量,采用双相滴定法测定白芥子碱含量。 以外源激素诱发的去势小鼠前列腺增生为动物模型,初步确定了白芥子抑制前列腺增生的有效部位和有效物质。 从毛细血管的通透性、肉芽组织增生、酸性磷酸酶活性、5α-还原酶活性等方面,探讨白芥子主要化学成分抑制前列腺增生的作用机理。
【Abstract】 This paper is a study on the extraction, separation, and content determination of all kinds of chemical components in seeds of Brassica alba. It also discusses the pharmacological effect and their mechanisms on BPH. The experimental methods and results are as follows.First, decide the optimal extract conditions of all kinds of extract.Second, seperate sinalbin, sinapine, 3 -sitosterol, from extract of Brassica alba’s seeds.Third, use HPLC to measure the content of P -sitosterol; use GC and GC-MS to analyze and measure the component and content of fatty acid in ether extract of Brassica alba’s seeds , the component and content of amino acid in seeds of Brassica alba are also analyzed and measured.Fourth, the medical effect of extracts and monomers obtained are filtrated using animal experiment of castrated mice prostatic hyperplasia induced by exterior hormone, several extracts and monomers which can significantly inhibit mice prostatic hyperplasia are found.Finally, by referring to the penetrability of capillary vesseK the hyperplasia of flesh bud and the activity of ACP and 5 a -reductase etc, the pharmacological mechanism of components inhibiting BPH are discussed.
