尼罗罗非鱼胰岛显微和亚显微结构的研究

【摘要】 本文应用光镜、电镜及特殊染色等技术对尼罗罗非鱼(Tilapia nilotica)的胰岛及其细胞类型和形态进行了详细的研究。 尼罗罗非鱼的胰脏与肝脏混合存在,属于肝胰脏。胰岛则散布在胰腺内,无固定的形态,有的胰岛只有几个胰岛细胞,胰岛细胞的数量与类型也因胰岛的大小而异。在H-E染色切片中,很容易将胰腺泡和胰岛区分开,前者染色深,数量多;后者染色淡,数量少。用Gomori醛复红特殊染色法,见尼罗罗非鱼胰岛有三种细胞:A细胞胞质中有黄色颗粒;B细胞胞质中有紫红色的颗粒;D细胞胞质内有浅绿色或淡灰色的颗粒。 电镜下,胰腺泡细胞内有非常发达的粗面内质网,排列呈板层状,靠近腺泡腔的胞质中有许多大而圆的酶原颗粒。胰岛细胞内的粗面内质网明显不如胰腺泡细胞的发达,其它细胞器也比较少;分泌颗粒小,外有界膜包被,颗粒大小、形态不完全相同,但其中央都有一个电子密度不等的芯。B细胞的分泌颗 王晓丽 尼罗罗非鱼胰岛显微和亚显微结构的研究13粒较大,芯的形态多样,电子密度差异大,芯与界膜之间常有较大的空隙;A细胞分泌颗粒形态不规则,芯的大小不等,界膜与芯之间无空隙;D细胞分泌颗粒较多,芯的电子密度较低,界膜与芯之间无空隙,此外,胞质内可见较大的线粒体。

【Abstract】 The type and morphology of the pancreas islet as well as its cells in Tilapia Nilotica were studied in detail by using the microscope, electron microscope and special staining techniques.The pancreas in Tilapia Nilotica which combine with liver, belongs to hepatopancreas. The pancreas islet, without fixed form, scatters in the pancreas. The number and type of pancreatic endocrine cells change following the size of pancreas islet, and some islets only have a few cells. We can distinguish clearly acinar cell of pancreas and pancreas islet by H-E staining, the former was dyed deep, the latter was dyed light. With the help of Gomori aldehyde fuchsine stain, we can find three kinds cells in pancreas islet. There are yellow granules in A cell, purplish red granules in B cell and green or light gray granules in D cell.There are very developed rough surfaced endoplasmic reticulums (RER) which was lamellar arrange in cytoplasm of acinar cells and there are much zymogen granules in cytoplasm where approach the lumen of acinus under the electron microscope. The RER in islet cells are not developed than in acinar cells and other organelles are scare to be seen too. Endocrine granules are small and packeted withmembrane. The size and type of granules are not entirely similar, but they all have a core with differ in density. Secretic granules in B cell are fairly big , cores are various. There is fairly big gap between the core and the limiting membrane; the center of secretic granules in A cell exist compact core and there is no gap between the core and the limiting membrane; Secretic granules in D cell are too much, electron density of granules is lower, and there is no gap between the core and the limiting membrane, what’s more, fairly big chondriosome can be seen in the cell.
