用现代分离技术提取抗癌活性成分喜树碱的研究

【摘要】 本文报告了开展利用现代技术(超临界二氧化碳流体萃取、大孔树脂分离)提取抗癌活性成分喜树碱的研究工作。文章首先综述了喜树碱的国内外研究进展,包括喜树的形态及资源分布、喜树的主要化学成分、喜树碱的结构与理化性质、喜树碱的结构和抗癌活性的关系、喜树碱的抗癌机理、喜树碱的修饰、喜树碱的提取研究等,并对超临界萃取技术、大孔树脂分离等几种中草药现代分离提取技术进行了详细的介绍。 本文研究了采用高压液相色谱法测定喜树碱的方法条件,获得了测定的回归方程及最小检出限等一系列结果。以此方法测定了采用超临界CO2萃取技术得到的喜树果萃取物中的喜树碱的含量,实验重现性良好。发现了喜树碱的闭环形式对光稳定和热不稳定性的实验事实。 本文优化了超临界CO2流体萃取喜树碱的实验条件和大孔树脂分离喜树碱的实验条件,获得超临界CO2流体萃取喜树碱的最优实验条件为:以0.6ml/g的比例直接向萃取原料添加乙醇作为提携剂,压力为35MPa,温度55℃,CO2用量80Kg(CO2)/Kg(原料);大孔树脂分离喜树碱的最佳实验条件为:氯仿/乙醇(1:1)作解吸液,解吸液PH值为3,试液上柱流速1.0ml/min。 本文首次采用超临界CO2流体萃取技术获得喜树果挥发油,获得无色透明的与喜树果粉香味完全一致的油状物,得油率为1.60%,其得油率是采用水蒸气蒸馏法获得的挥发油的4.6倍。并首次证实在喜树果挥发油中存在1,2-丙二醇、吡啶、1,3-丁二醇、2,3-丁二醇、己内酰胺、十六酸、十六酸乙酯、亚油酸、亚麻酸甲酯、亚麻酸乙酯、十八酸乙酯等物质。 本文为寻求一快速、廉价、低毒、少污染的喜树碱工业生产方法作了有意义的探索。

【Abstract】 The work used modern analysis technology(Supercritical CO2 extraction, macroreticular resin) on extraction antitumor activity compound CPT.At first, development of CPT in country and oversea was introduced, including conformation and resource distributing,main chemical ingredient,structure and physical chemistry property, relationship between structure and anti-tumor activity mechanism, CPT of modification, CPT of extraction etc. Particnlarly modern extraction and seperation technology of Chinese traditional madicine,such as SCF and macroreticular resin were introduced.Condition of Determination CPT content by HPLC was emphasized. The results rum out to be an regressive equation. CPT content in camptotheca acuminata fruit extracted by SCF was determined and gained good recurrently.The results showed close loop form of CPT was stable to light but instable to heat. The experiment condition of extraction of CPT by SCF and separation by macroporous resin were optimized.Optimization experiment condition by SCF find out to be:pressure 35Mpa,Tempreture 55 癈,dosage CC>2 80kg(co2/kg(materiai)-The optimization condition by macvoporous resin is:chloroform/alcohol(l:l), PH=3, velocity l.Oml/min.Extration of volatile oil from camptotheca acuminata fruit by SCF was introduced for the first time.Colourless clarity oil which scent identical to camptotheca acuminata fruit was gained.Ratio was 1.60%, 4.6 time as that by vapor distillation.Rapid cheapness CPT’S industrialization produce was also low poison, little pollution and effective method for explored in this paper.
