PVA-SbQ固定叶绿体及其在检测除草剂中的应用研究

【摘要】 聚乙烯醇-苯乙烯吡啶(PVA-SbQ)是一种新的光敏聚合物,在紫外光诱导下形成大分子网状结构,电镜照片显示PVA-SbQ固定的新鲜菠菜叶绿体-20℃贮存6月后,内、外膜结构完整,类囊体膜排列整齐,还原DPIP的能力保留50%。 叶绿体可以抵抗外界和内部胁迫,体内酶系可以降解超氧离子和H2O2,而除草剂可以影响叶绿体这种能力。将叶绿体固定膜固定在铂电极上,制成生物传感器,在含0.03%[H2O2]的50mMpH7.4 Tris-HCl缓冲液中,25℃条件下用示差脉冲伏安法检测除草剂。百草枯、敌草隆、莠去津、扑草净和阿特拉津的工作曲线浓度范围分别为0.0014~0.028、0.019~0.19、0.12~1.2、0.20~1.0和0.24~1.2 ppm。 此生物传感器不仅可以检测除草剂,还可以检测重金属离子和杀虫剂。应用该传感器可进行饮用水健康标准评价。

【Abstract】 Polyvinylalcohol bearing styrylpyridinium group is a new photo-induced cross-linking polymer. Ultraviolet can induced it to form network. Ultra-thin section photographs showed the intactness and orderly arrangement of the inner and outer membranes of the thylakoid entrapped in PVA-SbQ, after stored under -20 C for half a year. The activity of the membrane remained over 50%.Chloroplast has the ability to defense outer and inner stresses because it’s inner enzymes system can scavenge superoxygen radical and hydrogen peroxide. And herbicides can affect this ability. Stick the immobilized membrane to Pt electrode surface. Using a differential pause voltammetry method working in 50mM Tris-HCl (pH 7.4) buffer(with 0.03 % H202) under 25*C, the concentration of working curves of paraquat, diuron,ametryn,prometryn and atrazine ranged 0.0014-0.028, 0.019-0.19, 0.12-1.20, 0.20-1.0, 0.24-1.20 ppm respectively.This chloroplast biosensor can be used not only to detect herbicides, heavy metal ion and pesticides, but to evaluate the quality of drinking water.
