春性小麦抗穗发芽基因的转移及再生植株的培养

【摘要】 本研究应用PDS—1000/He气动式基因枪对三个春性小麦品种的幼胚、幼胚愈伤组织、幼穗进行了抗穗发芽基因PHS—r的转移,用抗除草剂的Bar基因作为筛选标记与其共转化。结果表明:在幼胚诱导愈伤组织的前2周用0.5mg/L ABA处理可有效抑制出芽率并提高愈伤组织的再生能力;0.2mg/L NAA和0.8mg/L IAA可促进幼苗生根;同时进行愈伤组织的筛选和分化培养可缩短接种到再生植株形成的历期;三种受体材料的再生率为幼胚>幼胚愈伤组织>幼穗。研究最终得到7株转化的再生植株,经PPT涂抹检测和PCR—电泳检测有4株再生小麦植株含有目的基因,转化率为0.37%。

Seven green plants were regenerr from immature embryos and calli of immature embryos. There are four pkzhiet containing the Pre-harvesting sprouting resistant gene in the genomc b2 test and PPT-test. The transforming ratio is 0.37%.
