黄瓜感染枯萎病菌后生理生化变化及其与抗病性关系的研究

【摘要】 本试验测定了黄瓜不同抗病性品种感染枯萎病菌和用病原菌毒素粗提液处理后植株体内的某些保护酶(POD、SOD和CAT)、抗性物质(还原糖、Vc、可溶性蛋白)、MDA含量和电导率的动态变化,并分析了这些生理生化变化与植物抗病性的相互关系,同时还对几种不同化合物对黄瓜枯萎病菌毒素的钝化作用进行了初步研究,结果表明: 1.无论是在黄瓜发芽期还是幼苗期,接菌处理和毒素粗提液处理后,其根部和叶部的SOD、POD活性与抗病性密切相关。抗病品种SOD、POD活性增加比率均明显高于感病品种。 2.无论是在黄瓜发芽期还是幼苗期,接菌处理和毒素粗提液处理后,SOD、POD、CAT、MDA含量和电导率处理比对照增减比率的变异幅度,抗病品种均显著低于感病品种。 3.在黄瓜幼苗期,无论是接菌处理还是毒素粗提液处理,还原糖含量、可溶性蛋白含量和Vc含量的变化都与抗病性密切相关。一般表现为抗病品种的含量高于感病品种。 4.黄瓜植株受病菌侵染后,根部保护酶活性(POD、SOD和CAT)、抗性物质(还原糖、Vc、可溶性蛋白)、MDA含量和电导率的变化均比叶部大。 5.黄瓜植株受病菌侵染后产生毒素是其主要致病因子之一。其中50%的毒素粗提液处理后MDA含量和膜透性相对较高,细胞膜受损伤程度相对较重。 6.黄瓜植株接菌处理和毒素粗提液处理均能引起不同抗病类型品种SOD、POD同工酶谱强度的变化。处理48小时后,抗病品种SOD、POD同工酶谱比对照的增强幅度明显高于感病品种。 7.KMnO4、CuSO4、FeCL3对黄瓜毒素粗提液均有一定的钝化作用,且不同浓度其钝化程度不同。以KMnO4的作用效果最明显。
【Abstract】 Studies on the relationships of some physilogical-chemical changes and disease resistance in cucumber infected with Fusarium wilt disease Abstract The activities of superoxide dismutase(SOD) peroxidease (POD) and catalyse(CAT)、 MDA content. relative electrical conductivity、 sugar、 water -soluble protein and Vc content in germinated seeds, cotyledons inoculated with Fusarium oxysporum and treated with crude extract of Fusarium wilt pathogen of cucumber cultivars with different resistance to Fusarium wilt of cucumber were measured. Meanwhile, some compounds inactivating the phytotoxicity to cucumber were studied.The results showed as follows: 1. POD and SOD activities have close correlation with the resistance of cucumber cultivars both in germinated seeds and in roots or leaves of cucumber in cotyledons. 2. Resistant cultivars, compared with susceptible cultivars, display low range of SOD、 POD CAT activity and membrane lipid peroxidation and relative electrical conductivity between treatment and control,. 3. Sugar, soluble protein and Vc content have close correlation with the resistance of cucumber cultivars in roots or leaves of cucumber in cotyledons . Resistant cultivars, compared with susceptible cultivars,have high sugar, soluble protein and Vc content. 4. Changes of the activities of superoxide dismutase(SOD). peroxidease (POD) and catalyse(CAT)、 MDA content、 relative electrical conductivity、. sugar、 water -soluble protein and Vc content in roots are more distinct than those in leaves when cucumber was inoculated with Fusarium oxysporum. 5. Toxin produced after cucumber inoculated with Fusariuni wilt pathogen was one of main pathogenic factors.There were high membrane lipid peroxidation and relative electrical conductivity after cucumber is treated with 50% toxin crude extract. 6. There were changes in the SOD and POD isozyme bands of inoculation and toxin treatment among different resistant cucumber cultivars . Isozyme bands of resistant cultivars are stronger than susceptible cultivars, compared with respective control. 7. KMnO4. CuSO4. FeCL3 can reduce the phytotoxicity of crude extract of cucumber wilt disease. Among these compounds, KMnO4 has best effect.
