草苁蓉对亚急性衰老大鼠抗衰老作用的实验研究

【摘要】 目的:探讨草苁蓉对亚急性衰老大鼠脑自由基、超微结构及组织化学的影响。方法:Wistar系大鼠随机分正常组、模型组(D—半乳糖48mg/kg/d)、人参皂甙组(100mg/kg.ig,D—半乳糖48mg/kg/d)、草苁蓉大剂量组(200mg/kg.ig,D—半乳糖48mg/kg/d)、草苁蓉中剂量组(150mg/kg.ig,D—半乳糖48mg/kg/d)、草苁蓉小剂量组(100mg/kg.ig,D—半乳糖48mg/kg/d),连续40天,取大脑皮质进行生化测定SOD、MDA、MAO;组化观察LDH、SDH、LPF及透射电镜观察。结果:注射D—半乳糖后,脑组织SOD活性降低(P<0.05),MDA含量及MAO活性增高(P<0.05),草苁蓉组可提高脑组织SOD活性(P<0.05),降低MDA含量(P<0.05),抑制MAO活性(P<0.05);组织化学及超微结构观察表明,注射D—半乳糖后,SDH活性下降、LDH活性增高,脂褐素、溶酶体大小形态不规则,线粒体肿胀、变性、嵴断裂、空泡样变,核膜形态不规则等衰老改变。草苁蓉可使上述变性程度减轻。结论:草苁蓉可延缓D—半乳糖所致的大鼠衰老,抑制脂质过氧化物的形成,提高抗氧化酶的活性,这可能是其抗衰老的作用机理之一。

【Abstract】 Objective: To study the effect of Boschniakia rossica to intracephalic free radical, ultrastructure and histochemistry of subacute senile rats. Method: To divide wistar rats randomizly into normal group, model group (D-galactose, 48mg/kg/d), ginsenoside group (lOOmg/kg.ig, D-galactose 48mg/kg/d), large dose group of Boschniakia rossica (200mgIkg.ig, D- galactose 48mg/kg/d), middle dose group of Boschniakia rossica (1 5Omg/kg.ig, D-galactose 48mglkgld). small dose group of Boschniakia rossica (1 00mg/kg. ig, D-galactose 48mg/kg/d), continuously 40 days, and biochemically analyse SOD, MDA, MAO and observe LDH, SDH, LPF and transmission electron microscope. Results: After injection of D-galactose, SOD of brain tissue was decreased (P<0.05). MDA content and MAO 2 activity were increased (P<O.05). Boschniakia rossica can increase SOD activity in brain tissue (P?O.05), decrease the content of MDA (P<O.05), restrain the MAO activity (P<O.05). According the observation of biochemistry and ultrastructure, after injection of D-galactose, SDH activity was decreased, LDH activity was increased, the shape of Lipofuscin and lysosome were irregular, mitochondrion swelling and become deformed, crest breaking and vacuolar degeneration, and shape of nuclear membrane was irregular. Boschniakia rossica alleviated denatured degree above. Conclusion: Boschniakia rossica can retard rats?senility made by D-galactose, restrain the form of lipid peroxid, increase the activity of antioxidase, these may be one of the mechanism action of anti senility.
