刺五加有效成分的提取、纯化及利用

【摘要】 本文依据刺五加中各成分的性质差异,应用不同的提取、纯化及分析方法,分别对各有效成分进行研究,总结出一条经济高效的工艺路线,实现了刺五加中异嗪皮定、刺五加甙、刺五加多糖的有效分离,为刺五加类药物形成系列产品奠定了基础。 1 首次运用超临界二氧化碳萃取技术提取刺五加中异嗪皮定,经高效液相分析,产品中异嗪皮定的含量达60%以上,收率达0.5281‰(半成品重/原料干重)。 2 刺五加生药粉经超临界技术选择性萃取后,其中所含刺五加甙和刺五加多糖等极性较强物质不受影响,因而用超临界萃取后的废料进行刺五加总甙和多糖提取研究,并对超声波提取和常温提取刺五加甙进行对比研究,应用大孔吸附树脂技术对刺五加甙进行纯化,经高效液相分析,所得产品中刺五加甙含量达60%以上,收率达0.4136%(半成品重/原料干重)。 3 树脂吸附过的刺五加原液进一步处理得刺五加多糖产品,经紫外分析,收率达0.5071%,纯度达70.10%。

【Abstract】 AbstractAccording to the different property of every composition in Acanthopanax senticosus, this paper applied different methods of extract, depuration and analysis and researched into these composition. As a result, a economical and efficient technical route is found. Through this route isofraxidin,cleutheroside and polysaccharides in Aanthopanax senticosus are separated effectively, and this study lay a solid foundation for the form of a series of products.(1)Isofraxidin was exL~~.~zd from the Acantl2opanax senticosus roots using supercritical CO2. The product was analyzed by HPLC. As a result, the content of isofraxidin is 60% and the yield of isofraxidin is 0.5281%.(2)The content of eleutheroside and polysaccharides was not influenced in Medicinal materials which have been extracted selectively by supercritical C02, So we began to study eleutheroside and polysaccharides in flotsam. The contradistinctive study between Ultra Sonic Extraction and Routine Extraction was done , and the resin was used in the depuration. The product was analyzed by HPLC. As a result, the content of eleutheroside is 60% and the yield of eleutheroside is 0.4 136%.(3)The extracted liquid which has been got rid of eleutheroside was managed more by chemic methods. The product was analyzed by LJV. As a result, the content of polysaccharides is 70.10% and the yield of polysaccharides is 0.5071%.
