金水六君煎质量标准研究

【摘要】 金水六君煎出自明代张景岳《景岳全书》,该方由半夏、陈皮、当归、熟地、茯苓、炙甘草、生姜共七味中药材搭配组成,主治肺肾阴虚,水泛为痰,咳嗽呕恶,喘逆多痰,痰带咸味等症。暨南大学医学院沈英森、赵长鹰医师等人对金水六君煎作了长期的临床(204例)与动物实验研究,结果表明该方治疗慢性支气管炎有良好的疗效。为使该方能够推向市场,必须建立质量标准,本文运用现代色谱和光谱手段对其质量控制方法进行了研究。首先,我们使用薄层色谱和薄层扫描法通过薄层层析图谱、光谱扫描图谱和薄层扫描图谱等指标对金水六君煎和方中各单味药材进行了定性鉴别。其次,我们用高效液相色谱法测定了金水六君煎中橙皮苷和阿魏酸的含量,并对阿魏酸的稳定性和色谱优化条件进行了研究。由于橙皮苷属于黄酮类化合物,陈皮和甘草等药材中都含有黄酮类化合物,所以还用紫外分光光度法测定了金水六君煎中总黄酮含量。从而建立起金水六君煎的一整套定性、定量质量标准。最后,我们对陈皮的有效成分橙皮苷进行了动物实验研究,结果表明橙皮苷有较强的祛痰作用,能提高慢性支气管炎模型大鼠肺泡灌洗液中的总磷脂含量,提高肺表面活性物质的量和活性,促进纤毛的生长和运动,对慢性支气管炎有较强的药理活性,是金水六君煎的活性有效成分,这些研究表明我们建立起来的以金水六君煎中橙皮苷含量为主要质量控制指标的质量标准是行之有效的。

【Abstract】 JinShuiLiu.JunJian come from book of Jingyue Complete Works written by Zhang Jingyue in Ming Dynasty, it is made of Rhizoma Pinelliae.. Pericarpium Citri Reticulatae, Radix Angelicae Sinensis, Radix Glycyrrbizae, Poria, Radix Rehmanniae Preparata and Rhizoma Zingiberis Recens. It mainly be used to cure yin-deficiency of lung and kidney, transmission of dryness and dampness into phlegm, cough, vomiting, nausea, syndrome characterized by dyspnea with phlegm, phlegm with salty taste and so on. Shen Yingsen and Zhao Changying in Medical College of Jinan University had done long-term research of clinic (204 cases) and animal experiment, the result of research indicated that JinShuiLiuiunJian had well therapeutic effect to chronic bronchitis. In order to marketing JinShuiLiuJunJian, its quality standard must be built. This paper researched its quality control method by modem chromatographic and spectrographic instrument. The first, JinShuiLiuiun -Jian and its single medicinal materials were qualitatively identified by thin layer chromatography and thin layer chromatographic scanning through quality index of thin layer chromatogram thin layer scanning chromatogram and ultraviolet scanning spectrogram etc., The second, the content of hesperidin and ferulic acid in JinShuiLiuiunJian were determined by high performance liquid chromatography and the stability and optimize chromatographic determination conditions of ferulic acid were researched. Because hesperidin belongs to flavone compound and that flavone compound had been found in Pericarpium Curl Reticulatae and Radix Rehmanniae Preparata, the content of total flavone compounds in JinShuiLiuJunJian was determined by ultraviolet spect2rophotometry. So the quantitative and qualitative quality standard was built. The last, the hesperidin’s animal experiment was researched which was the effectual component of Pericarpium Citri Reticulatae. The result of animal experiment indicated that hesperidin had strong pectroral ability, could improve the content of total phospholipid in I3ALF of big rat of chronic bronchitis model to improve the activity and quantity of pulmonary surfactant,could promote movement and growth of cilium, was the active component of JinShuiLiuJunJian. Those researches proved that our quality standard was effective which primary quality control index was content determination of hesperidin in JinShuiLiuJunJian.
