益气通络方对大鼠前脑缺血后海马迟发性神经元死亡保护作用的研究

STUDIES ON EFFECTS OF YQTLD ON DELAYED NEURONAL DEATH OF HIPPOCAMPUS OF TRANSIENT GLOBAL FOREBRAIN ISCHEMIA OF RAT

【摘要】 目的 探讨益气通络方对大鼠短暂性脑缺血后海马迟发性神经元死亡的保护作用及其机制。方法 用Pulsinelli四血管闭塞法制作全脑缺血模型。实验动物随机分为(1)假手术对照组;(2)脑缺血组;(3)生理盐水+脑缺血组(4)益气通络方+脑缺血组(5)脑缺血+生理盐水组和(6)脑缺血+益气通络方组。3天后进行HE染色、TUNEL染色和免疫组化。结果(一)HE染色,光镜下观察CA1区组织病理学变化,并用显微测微尺测量CA1区存活的锥体细胞密度(锥体细胞的个数/毫米):(1)假手术对照组245.0±14.0;(2)脑缺血组9.5±2.5;(3)生理盐水+组脑缺血10.5±3.0;(4)益气通络方+脑缺血组114.5±19.0;(5)脑缺血+生理盐水组9.0±3.5;(6)脑缺血+益气通络方组10.0±3.5。(二)TUNEL染色:光镜下假手术组未见阳性结果。缺血再灌流3天后,CA1区大部分锥体神经元TUNEL染色阳性,生理盐水组与其无明显差别。益气通络方+缺血组CA1区TUNEL染色阳性的锥体神经元明显减少,存活细胞增多,每组CA1区TUNEL染色阳性细胞数/毫米:(1)脑缺血组121.0±9.5;(2)生理盐水组124.0±11.5;(3)益气通络方组24.5±3.0;(三)免疫组织化学结果显示,caspase免疫反应阳性产物呈褐色颗粒状,缺血组脑片CA1区染色阳性,呈深褐色,益气通络方+缺血组脑片CA1区染色也可见阳性染色,但颜色明显变浅。假手术组脑片CA1区染色阴性。结论 (一)益气通络方能够显著降低大鼠短暂性脑缺血后海马迟发性神经元死亡,抑制细胞凋亡可能是其机制之一。(二)益气通络方可以下调大鼠短暂性脑缺血后海马神经元caspase-3的表达,从而提示该方可能通过此途径抑制脑缺血后海马迟发性神经元凋亡,这可能是其临床疗效的机制之一。

【Abstract】 Objective: To investigate effect of Yiqi Tongluo Decoction (YQTLD), a traditional Chinese medicinal prescription, on delayed neuronal death of hippocampus of transient global forebrain ischemia in rat. Methods: Transient forebrain ischemia was produced using published modifications of the four-vessel occlusion (4-VO) method and it triggered the delayed death of CAl neurons in the hippocampus. YQTLD was given orally to rats. Then, we studied the effect of YQTLD on cell survival and DNA fragmentation in the hippocampus in a rat model of transient global ischemia and on expression of caspase-3P20 in CAl neurons, indicative of apoptosis. Results: (1)oral administration of YQTLD has effect on cell survival. However, YQTLD significantly intensified the number of survival neurons. (2) YQTLD markedly attenuated the number of TUNEL positive neurons. (3) the expression of caspase-3P20 decreased in CAl nuerons after transient forebrain ischemia of oral administration rats of YQTLD. Conclusion: The present results suggested that YQTLD given orally prevented the necorsis and apoptosis and prevented caspase-3P20, the production of catalytically active caspase-3, which might account for the enhancing effect of YQTLD on cell survival by preventing caspae-3 way.
