猪囊尾蚴的生化代谢规模和药物作用机理

【摘要】 本研究通过测定发育过程中猪囊尾蚴及抗囊药物作用下猪囊尾蚴的多种生化代谢指标,阐明了猪囊尾蚴的基本生化代谢规律和药物作用机理,为猪囊尾蚴病的科学防治提供了重要的实验和理论依据。 从发育过程中的猪囊尾蚴检测到参与物质吸收转运的三种ATPase、ALP,糖类物质代谢的GLc、HK、MDH、PK、LDH、LAC、G6PDH,脂类物质代谢的TG,氨基酸代谢的GOT、GPT、GDH、UN,嘌呤代谢的XOD、UA。发现囊壁HK、PK、MDH、Na+K+—ATPase活性及GLc含量、头颈节Ca2+—ATPase活性随虫体由未成熟期进入成熟期而升高,表明虫体存在着类似于其它蠕虫的物质转运、糖有氧分解、糖无氧分解、磷酸戊糖途径、氨基酸代谢等生化代谢通路,并且随着虫体生长发育,膜转运功能加强,由HK催化GLc进入代谢的通路活跃,由PEP经PK、LDH催化生成LAC的支路和经MDH等催化进入逆向三羧酸循环的支路均加强。 实验结果表明,三种药物NX0、NX1、NX2对体外培养六钩蚴、未成熟猪囊尾蚴、成熟期猪囊尾蚴生化代谢均有一定作用,在成熟期NX0、NX1对猪囊尾蚴生化代谢的影响相似,即首先使HK、MDH活性及GLc含量降低,PK、LDH活性及LAC含量升高,于后期各代谢系统均受影响,酶活性下降,代谢产物蓄积。说明药物首先抑制MDH所在糖有氧分解通路,糖无氧分解过程代偿性增强,而后糖分解代谢全面抑制,致使虫体能量耗竭,直至死亡。药物对体内各时期猪囊尾蚴生化代谢的影响,根据疗效观察NX2无效,NX1优于NX0,NX1对未成熟期猪囊尾蚴的杀灭作用优于成熟期。

【Abstract】 By detecting a few biochemical indexes of C.cellulosae in the development and treated by drugs,the article studied principle of biochemical metabolism of C.cellulosae and mechanism of action of drugs and provided a theoretical basis for preventing and treatment of cysticercosis. Three kinds of ATPase and ALP wtiich join absorption and transport of substance, Glc HK. MDH. PK. LDH. LAC and G6PDH in glycometabolism,TG in lopid metabolism,GOT GPT、GDH and UN in amino acid metabolism and XOD and UA in purine metabolism were detected .Activity of HK.. PK MDH. NaKTPase and content of GLc of bladder wall and activity of Ca2TPase of scolex rised with the development of C.collulosae.The results showed that transport of substance,anaerobic glycolysis,aerobic glycotysis,phosphopentose pathway and amino acid metabolism were same as those in other helminths. With the development of C.cellulosae,fiinction of membrane transport,the pathways in which HK catalyzed GLc to metabolize and PEP formed LAC with the catalysis of PK and LDH and the pathway in which the parasite entered into reverse tricarboxylic acid cycle enhanced. The detection results of effects of drugs on biochemical metabolism of C.cellulosae cultured in vitro in different periods showed that NX NX1 and NX2 had some actions on oncospheres,immature and mature C.cellulosae and NX0 had the same effects against biochemical metabolism of C.cellulosae as NX1 did.First activity of MDH and content of GLc decreased and activities of PK and LDH and content of LAC increased. At the later stage ,alI the metabolism systems were effectd and activies of enzymes decreased and metabolite accumulated. The results showed that drugs inhibited the pathway of aerobic glycolysis and anaerobic glycolysis strengthened compensatorily and then the metabolism of glycolysis were inhited,which resulted in exhaustion of energy and death of C.cellulosea. According to the therapeutic effectiveness of dntgs,NX2 was ineffective and NX1 was better than NX0 and NX1 had better treatment agaist immature C.cellulosae than against mature C.cellulosae in vivo.
