番茄褐色皱纹果病毒分离物鉴定及进化分析

【摘要】 番茄褐色皱纹果病毒(tomato brown rugose fruit virus,ToBRFV)为帚状病毒科(Virgaviridae)烟草花叶病毒属(Tobamovirus)的RNA病毒。在内蒙古自治区某番茄大棚发现,感病番茄果实呈褐色斑块且皱缩。本研究利用特异性引物进行扩增获得560 bp特异性片段,经序列比对,与ToBRFV以色列分离株(MN882023.1)、墨西哥分离株(MN882024.1)的核苷酸同源性达98.7%。比对不同地区ToBRFV基因组,构建进化树,确认ToBRFV毒株所带病毒与北美分支亲缘性最高。本研究明确了病原体的分类地位及其与全球流行分离株的高度同源性,该发现为解析ToBRFV的分子进化特征、后续抗病基因资源挖掘及抗病机制研究提供关键理论依据和实验基础。

【Abstract】 Tomato brown rugose fruit virus(ToBRFV), a member of the genus Tobamovirus in the family Virgaviridae, is an RNA virus. It was discovered in a tomato greenhouse in Inner Mongolia Autonomous Region,where the infected tomato fruits showed brown patches and wrinkles. In this study, a 560 bp specific fragment was obtained by amplification using specific primers. Sequence comparison revealed that the nucleotide homology with the Israeli isolate MN882023.1 and the Mexican isolate MN882024.1 of ToBRFV was 98.7%. By comparing the genomes of ToBRFV from different regions and constructing an evolutionary tree,it was confirmed that the ToBRFV strain in this study was most closely related to the North American branch. This study clarified the taxonomic status of the pathogen and its high homology with globally prevalent isolates, providing a key theoretical basis and experimental foundation for understanding the molecular evolutionary characteristics of ToBRFV, as well as for the subsequent exploration of disease-resistant gene resources and the study of disease resistance mechanisms.
