鹦鹉热衣原体数字PCR检测方法的建立与应用

【摘要】 鹦鹉热衣原体(Chlamydia psittaci,C.psittaci)是一种引起呼吸道感染为主的人畜共患病原体,因无特异性临床症状,易漏诊误诊,可能导致重症,其危害性常被低估。本研究基于GenBank已发表鹦鹉热衣原体ompA基因(GenBank登录号:PQ511242.1)的核苷酸序列设计1对引物和探针,建立鹦鹉热数字PCR(digital PCR,dPCR)检测方法,优化反应体系。结果表明,dPCR的最佳引物和探针浓度分别为900 nmol/L和250 nmol/L;最佳的退火温度为56℃。该方法最低检测限为0.58 copies/μL,线性关系良好,与其他7种病原无交叉反应,具有特异性高、重复性好的特点。临床检测72份牛羊血液及阴道拭子样品,dPCR阳性检出率为19.04%,实时荧光定量PCR (real-time quantitative PCR,qPCR)阳性检出率为18.06%。本研究建立的dPCR检测方法比qPCR方法更加灵敏、高效,可用于鹦鹉热衣原体的定量检测。

【Abstract】 Chlamydia psittaci(C. psittaci) is a zoonotic pathogen that primarily causes respiratory tract infections. Due to the absence of specific clinical symptoms, it is prone to missed diagnosis and misdiagnosis, which may lead to severe illness, and its harmfulness is often underestimated. In this study, a pair of primers and a probe were designed based on the nucleotide sequence of the C. psittaci ompA gene(GenBank accession number: PQ511242.1) published in GenBank. A digital PCR(dPCR) detection method for C. psittaci was established, and the reaction system was optimized. The results showed that the optimal concentrations of primers and probe for dPCR were 900 nmol/L and 250 nmol/L, respectively, and the optimal annealing temperature was 56 ℃. The method achieved a lower limit of detection of 0.58 copies/μL, demonstrated a good linear relationship, and showed no cross-reaction with seven other pathogens, indicating high specificity and good reproducibility. In the clinical detection of 72 bovine and ovine blood and vaginal swab samples, the positive detection rate of dPCR was 19.04%, while that of real-time quantitative PCR(qPCR) was 18.06%. The dPCR method established in this study is more sensitive and efficient than the qPCR method and can be used for the quantitative detection of C. psittaci.
