气雾吸入CpG佐剂对新城疫弱毒苗免疫效果的影响
Immune-enhancing effect of a CpG adjuvant on a Newcastle disease attenuated vaccine administered by aerosol inhalation
【摘要】 为研究CpG序列对新城疫弱毒苗的免疫效果,本试验将75只7日龄SPF鸡随机分为5组,即空白对照组,疫苗对照组,高、中、低剂量雾化组。将不同剂量的pVAX-CpG10佐剂(100、50、25μg/只)与新城疫LaSota株弱毒疫苗混合,气雾免疫SPF鸡,并以无CpG10佐剂的新城疫LaSota株弱毒疫苗和雾化PBS作为对照组,通过每周检测免疫鸡的新城疫抗体效价,免疫后48 h肺组织CD3+、CD4+、CD8+ T淋巴细胞亚群的表达量,免疫后0、14 d外周血IgG、IL-6和IFN-γ的水平以及免疫后21 d免疫器官指数和肺部SIgA的分泌,评价CpG佐剂对新城疫弱毒苗的免疫增强效果。结果显示,中剂量雾化组在免后7、14 d新城疫抗体效价均显著高于免疫对照组(P<0.05);免疫后48 h中剂量雾化组肺组织CD3+、CD4+、CD8+T淋巴细胞亚群的表达量均极显著高于疫苗对照组(P<0.01);免疫后14 d中剂量雾化组IL-6和IFN-γ的水平均显著高于免疫对照组(P<0.05);免疫后28 d中剂量雾化组SIgA的含量极显著高于免疫对照组(P<0.01)。结果表明,优化的含有CpG序列的重组质粒pVAX1-CpG10不仅能有效提高新城疫弱毒疫苗的抗体效价,还能促进机体IL-6、IFN-γ以及SIgA的分泌,进而增强机体免疫力。
【Abstract】 In order to study the immune-enhancing effect of the recombinant plasmid pVAX1-CpG10 containing CpG sequence on Newcastle disease attenuated vaccine,75 seven-day-old SPF chickens were randomly divided into 5 groups,namely blank control group,vaccine control group,high-,medium-and low-dose nebulization group,among which the high-,medium-,and low-dose nebulization groups received the Newcastle disease LaSota strain attenuated vaccine nebulized with different doses of the CpG10 adjuvant(100,50,and 25 μg/dose,respectively).The vaccine control group was nebulized with the attenuated Newcastle disease LaSota strain without CpG10 adjuvant,and the blank control group was nebulized with PBS solution,The expression levels of CD3~+,CD4~+,and CD8~+ T lymphocyte subsets in lung tissue at 48 h post-immunization,the levels of IgG,IL-6 and IFN-γ in peripheral blood at 0 and 14 d post-immunization,and the immune organ index and SIgA levels in the lungs at 21 d were measured separately to evaluate the immune-enhancing effect of CpG adjuvant on attenuated Newcastle disease vaccine.The results showed that the titers of Newcastle disease antibody in the medium-dose nebulization group were significantly higher than those in the immunized control group at 7 and 14 d after immunization(P<0.05).The levels of CD3~+,CD4~+,and CD8~+ T-lymphocyte subsets in the lung tissue of the medium-dose nebulization group were significantly higher than those in the vaccine control group at 48 h after immunization(P<0.01).The levels of IL-6 and IFN-γ in the medium-dose nebulization group were significantly higher than those in the control group on the 14th day after immunization(P<0.05).The content of SIgA in the medium-dose nebulizer group was significantly higher than that in the immunized control group at 28 d after immunization(P<0.01).The results showed that the optimized recombinant plasmid pVAX1-CpG10 containing CpG sequence not only effectively increases the antibody titer of Newcastle disease attenuated vaccine,but also promotes the secretion of IL-6,IFN-γ and SIgA,thereby enhancing immune responses.
