硒代蛋氨酸对呕吐毒素致兔十二指肠损伤的干预作用
Intervention Effect of Selenomethionine on Duodenal Injury Caused by Deoxynivalenol in Rabbits
【摘要】 为了探究硒代蛋氨酸(Selenomethionine,SeMet)对呕吐毒素(Deoxynivalenol,DON)致兔十二指肠损伤的缓解作用。将40只35日龄兔随机分为4组(CON组、DON组、SeMet组和DON+SeMet组)。从试验第1天开始,SeMet组和DON+SeMet组每天饲喂0.2 mg/kg SeMet,持续21 d。第15天开始DON组和DON+SeMet组每天灌胃0.5 mg/kg的DON,持续7 d。攻毒结束后,采集兔十二指肠组织,通过HE、PAS染色和扫描电镜观察绒毛、隐窝形态、杯状细胞数量和超微结构变化,通过qPCR和ELISA分析组织相关基因和蛋白的表达水平。结果显示,与CON组相比,DON组十二指肠隐窝深度显著增加,绒毛高度/隐窝深度显著降低;扫描电镜观察绒毛表面呈颗粒状,侧面结构显示微绒毛排列不整齐,微皱褶不规则;DON组十二指肠组织内IL-6的mRNA表达水平显著升高,IL-6、IL-1β和TNF-α含量显著升高,IL-10含量显著降低。与DON组相比,DON+SeMet组绒毛高度和绒毛高度与隐窝深度之比显著提高,隐窝深度显著降低,杯状细胞数量显著增加,微绒毛排列相对整齐,微皱褶规则,十二指肠组织内IL-6的mRNA表达水平显著降低,IL-6、IL-1β和TNF-α蛋白水平显著降低,IL-10的含量显著增加。试验证实,在兔子日料中添加0.2 mg/kg SeMet在一定程度上可改善DON诱导的兔子十二指肠黏膜损伤。
【Abstract】 To investigate the protective effect of selenomethionine(SeMet) against deoxynivalenol(DON)-induced duodenal injury in rabbits.Forty 35-day-old rabbits were randomly and equally divided into four groups(blank control group,DON group,SeMet,DON+SeMet group).Starting from day 1 of the experiment,the SeMet and DON+SeMet groups were fed 0.2 mg/kg SeMet daily for 21 days.From day 15,the DON and DON+SeMet groups were intragastrically administered 0.5 mg/kg body weight of DON daily for 7 days.Duodenal tissues were collected from the rabbits,Observed by using HE staining,PAS staining,and scanning electron microscopy(SEM).The expression levels of related genes and proteins in the tissues were analyzed by qPCR and ELISA.Compared with the CON group,the DON group showed no significant difference in villus height,but a significant increase in crypt depth and a significant decrease in the height-to-crypt depth ratio.SEM observations revealed a granular surface of the villus,with disorganized microvilli arrangement and irregular microplicae on the lateral structures.In the DON group,the mRNA expression level of IL-6 in the duodenal tissue was significantly increased,the levels of IL-6,IL-1β,and TNF-α were significantly elevated,and the level of IL-10 was significantly decreased.Compared with the DON group,the DON+SeMet group showed a significant increase in villus height and the villus height-to-crypt depth ratio,a significant decrease in crypt depth,and a significant increase in the number of goblet cells).The microvilli were relatively well-arranged,and the microplicae were regular.The mRNA expression level of IL-6 in the duodenal tissue was significantly decreased,the protein levels of IL-6,IL-1β,and TNF-αwere significantly reduced,and the level of IL-10 was significantly increased.The experiment have shown that dietary supplementation with SeMet can partially alleviate DON-induced duodenal mucosal injury.
