骨硬化蛋白抗体治疗骨质疏松症的研究进展
Research progress of sclerostin antibody in the treatment of osteoporosis
【摘要】 骨质疏松作为一种全身代谢性骨病,与骨形成、骨吸收平衡破坏有关。Wnt/β-catenin信号通路作为骨骼形成的重要调控通路,主要通过影响成骨细胞分化进程来实现。由骨细胞分泌的硬化蛋白能够干扰该通路的活性,从而抑制骨组织形成。近年来,硬化蛋白单克隆抗体作为治疗骨质疏松症的新型药物已逐渐受到临床关注,该制剂能够有效促进骨骼生成,提高骨密度,改善骨骼力学性能,同时减缓骨吸收速度,为临床治疗骨质疏松症提供了新方向。本文基于近年国内外研究成果,系统分析该类药物的作用原理、治疗效果及潜在风险,以期为骨质疏松诊治预防工作提供参考。
【Abstract】 As a systemic metabolic bone disease, osteoporosis is related to bone formation and bone resorption balance destruction. As an important regulatory pathway of bone formation, Wnt/β-catenin signaling pathway is mainly achieved by affecting the differentiation process of osteoblasts. Sclerostin secreted by bone cells can interfere with the activity of this pathway, thereby inhibiting bone tissue formation. In recent years, sclerostin monoclonal antibody, as a new drug for the treatment of osteoporosis, has gradually attracted clinical attention, and can effectively promote bone formation, increase bone mineral density, improve bone mechanical properties and slow down bone resorption, thus providing a new direction for clinical treatment of osteoporosis. Based on the research results at home and abroad in recent years, this paper systematically analyzes the action principle, therapeutic effect and potential risk of the drug, in order to provide reference for the diagnosis,treatment and prevention of osteoporosis.
