小檗碱通过腺苷酸活化蛋白激酶/哺乳动物雷帕霉素靶蛋白信号通路对糖尿病大鼠附睾组织保护作用的研究

【摘要】 目的 探讨小檗碱(BBR)通过腺苷酸活化蛋白激酶(AMPK)/哺乳动物雷帕霉素靶蛋白(mTOR)信号通路对DM大鼠附睾组织的影响。方法 将40只SD雄性大鼠随机分为正常对照(NC)组、高脂高糖(HFG)组、DM组、BBR组,每组10只。检测各组附睾检测精子质量,Western blot检测附睾组织中苄氯素(Beclin1)、微管相关蛋白轻链3(LC3)、泛素结合蛋白(p62)、磷酸化mTOR/mTOR(p??mTOR/mTOR)、p??AMPK/AMPK、IL??1β和白介素??1β前体(proIL??1β)蛋白表达,qRT??PCR检测m RNA表达,HE染色检测附睾组织病理状态。结果 与NC组比较,DM组精子畸形率和mTOR、p62 mRNA表达及p??mTOR/mTOR、p62、proIL??1β、IL??1β蛋白表达升高(P<0.05),精子密度和AMPK 、Beclin1 m RNA表达及p??AMPK/AMPK 、Beclin1 、LC3Ⅱ/LC3Ⅰ蛋白表达降低(P<0.05)。与DM组比较,BBR组精子畸形率和mTOR、p62 mRNA表达及p??mTOR/mTOR、p62、proIL??1β、IL??1β蛋白表达降低(P<0.05),精子密度和AMPK、Beclin1 mRNA表达及p??AMPK/AMPK、Beclin1、LC3Ⅱ/LC3Ⅰ蛋白表达升高(P<0.05)。结论 BBR可缓解DM大鼠附睾病理状态,调节其炎症和自噬水平,保护DM大鼠附睾,改善雄性生殖障碍,可能与AMPK/mTOR通路激活相关。

【Abstract】 Objective To investigate the protective effect of berberine(BBR) on epididymal tissue in diabeties mellitus(DM) rats through the adenylate activated protein kinase(AMPK)/mammalian target of rapamycin(mTOR) signaling pathway. Methods 40 SD male rats were randomly divided into normal(NC) group,high??fat and high??glucose(HFG) group,DM group and BBR group,with 10 rats in each group. The epididymis was detected for sperm quality in each group,and benzyl chloride(Beclin1),microtubule??associated protein light chain 3(LC3),ubiquitin??binding protein(p62),phosphorylated mTOR/mTOR(p?? mTOR/mTOR),p?? AMPK/AMPK,IL?? 1β and interleukin 1?? β precursor(proIL?? 1β) were detected by Western blot. Results Compared with NC group,the sperm abnormality rate and the expressions of mTOR,p62 mRNA,p??mTOR/mTOR,p62,proIL??1β and IL??1β increased(P<0. 05),and the sperm density,AMPK,Beclin1 m RNA and p ?? AMPK/AMPK,Beclin1,LC3 Ⅱ/LC3 Ⅰ proteins decreased in DM group(P<0. 05). Compared with DM group,BBR group had lower sperm abnormality rate,expressions of mTOR and p62 m RNA and p??mTOR/mTOR,p62,proIL??1β and IL??1β proteins(P<0. 05),higher sperm density,AMPK,Beclin1 m RNA and p??AMPK/AMPK,Beclin1,LC3Ⅱ/LC3Ⅰ proteins(P<0. 05). Conclusions BBR can alleviate the pathological state of the epididymis in diabetic rats,regulate its inflammation and autophagy levels. BBR demonstrates a protective effect on the epididymis in diabetic rats,ameliorating male reproductive dysfunction,potentially through the activation of the AMPK/mTOR pathway.
