【摘要】 目的：分析免疫组化染色技术检测程序性死亡配体1(PDL1)及免疫细胞病理分析在膀胱尿路上皮癌联合检测的意义。方法：选取2022年1月5日～2024年1月5日就诊于本院泌尿外科的患者158例，随机分为病例组和膀胱良性疾病组各79例。使用BenchmarkULTRA全自动免疫组化染色仪，检测PDL1表达，经细胞玻片免疫捕获技术分析CD3、CD4、CD8阳性细胞计数及亚群比例，评估调节性T细胞(Treg)浸润情况。结果：病例组PDL1表达水平及CD3、CD4、CD8阳性细胞浸润程度均高于膀胱良性疾病组(P<0.05)；病例组CD8~+/CD3~+比例高于CD4~+/CD3~+比例，PDL1表达与CD3阳性细胞浸润呈正相关。此外，病例组Treg浸润水平与PDL1表达正相关(r=0.58±0.07)，与CD3阳性细胞浸润负相关(r=-0.42±0.05)。BenchmarkULTRA检测的技术重复性优异，PDL1表达及CD3、CD4、CD8阳性细胞计数的平均一致性分别达96.57%、97.82%、97.04%、97.47%。结论：BenchmarkULTRA凭借标准化操作、高定量精度及高重复性，能稳定、精准地量化PDL1表达及免疫细胞浸润水平。更多还原

【Abstract】 Objective:To analyze the significance of combined detection of programmed death ligand 1(PDL1) by immunohistochemical staining and immunocytochemistry in bladder urothelial carcinoma.Methods:A total of 158 patients who were admitted to the Department of Urology of our hospital from January 5,2022 to January 5,2024 were randomly divided into case group and benign bladder disease group,with 79 cases in each group.The expression of PDL1 was detected by Benchmark ULTRA automatic immunohistochemical staining instrument.The CD3,CD4,CD8 positive cell count and subgroup ratio were analyzed by cell slide immune capture technique,and the infiltration of regulatory T cells(Treg) was evaluated.Results:The expression level of PDL1 and the infiltration degree of CD3,CD4 and CD8 positive cells in the case group were higher than those in the benign bladder disease group(P<0.05).The proportion of CD8~+/CD3~+ was higher than that of CD4~+/CD3~+ in the case group,and the expression of PDL1 was positively correlated with the infiltration of CD3 positive cells.In addition,the level of Treg infiltration in the case group was positively correlated with PDL1 expression(r=0.58±0.07) and negatively correlated with CD3 positive cell infiltration(r=-0.42±0.05).The repeatability of Benchmark ULTRA was excellent.The average consistency of PDL1 expression and CD3,CD4 and CD8 positive cell counts was 96.57%,97.82%,97.04% and 97.47%,respectively.Conclusion:With standardized operation,high quantitative accuracy and high repeatability,Benchmark ULTRA can stably and accurately quantify the expression of PDL1 and the level of immune cell infiltration.更多还原
