梨砧木中矮3号组培快繁体系的建立

【摘要】 利用无性系砧木嫁接繁殖是果树现代化种苗繁育的发展方向,组织培养是矮化砧木无性系生产的有效途径之一。梨矮化砧木中矮3号生根困难,目前仅以嫁接方式作中间砧使用,尚未有无性系自根苗,这严重制约了其推广与应用。以脱毒的中矮3号组培苗为试材,对组培苗进行分级培养,筛选增殖培养与生根培养合适的植物生长调节剂种类与配比。结果表明:单芽弱苗(苗高<2.0 cm)继代培养于添加2.5 mg/L 6-BA、0.2 mg/L NAA、2.0 mg/L GA3的MS培养基中,可获得高比率丛生芽;带有愈伤组织的丛生芽弱苗继代培养于添加1.0 mg/L 6-BA、0.2 mg/L NAA、2.0 mg/L GA3的MS培养基中,可获得高比率壮苗;壮苗(苗高≥2.0 cm)转接至添加3.0 mg/L IBA的NN69培养基中,经两步生根诱导,生根率在90%以上,不定根数在8个以上。建立了中矮3号的离体繁殖技术体系,为梨矮化砧木的无性系生产和商业化应用提供了技术支撑。

【Abstract】 Grafting using clonal rootstocks is the trend for modern propagation of nursery plant stocks in fruit trees. Tissue culture is one of the effective ways to produce clonal dwarfing rootstocks. The pear dwarfing rootstock‘Zhong’ai 3’ has difficulty producing adventitious roots. At present, it is only used as an intermediate rootstock through grafting, and there are no clonal self-rooted seedlings yet, which seriously restricts its popularization and application. Using the detoxified tissue culture seedlings of ‘Zhong’ai 3’ as test materials, the tissue culture seedlings were cultured by grading to screen the appropriate types and ratios of plant growth regulators for proliferation culture and rooting culture. The results showed that a high proportion of multiple-shoots was obtained when weak singleshoots(seedling height<2.0 cm) were sub-cultured in MS medium supplemented with 2.5 mg/L 6-BA, 0.2 mg/L NAA,and 2.0 mg/L GA3. The weak multiple-shoots with callus were sub-cultured in MS medium supplemented with 1.0 mg/L 6-BA, 0.2 mg/L NAA, and 2.0 mg/L GA3, and a high ratio of strong single-shoots was produced. Strong single-shoots(seedling height≥2.0 cm) were induced to root in NN69 medium supplemented with 3.0 mg/L IBA by a two-step rooting strategy. More than 90% of the plantlets produced at least 8 adventitious roots. This study has established the protocol for in vitro propagating ‘Zhong’ai 3’, which provides technical support for the clonal production and commercial application of pear dwarfing rootstocks.
