不同产地细辛对小鼠的亚急性毒性初探

【摘要】 目的 为不同产地细辛的亚急性毒性及安全性研究提供参考。方法 采用最大给药量法,将60只KM小鼠随机分为对照组(等体积生理盐水)及S1给药组(药材产地辽宁)、S2给药组(药材产地吉林)、S3给药组(药材产地黑龙江)、S4给药组(药材产地湖北)、S5给药组(产地山东),各10只,雌雄各半,分别灌胃相应药液或生理盐水(灌胃体积4 g/kg),每日2次,持续14 d。观察小鼠一般状态、死亡情况及体质量变化情况;给药后14 d,取小鼠主要器官(肝、肺、肾)组织,采用苏木精-伊红(HE)染色,观察病理形态并进行病理学评分。结果 与对照组比较,各给药组小鼠表现出不同程度的毒性作用,均出现自发活动增加、暴躁乱蹿等行为异常,且陆续出现死亡,各给药组雌性小鼠死亡率高于雄性小鼠,且S3给药组和S4给药组小鼠总死亡率(40.00%)明显高于其他组;给药期间,整体上各组雄性小鼠体质量高于雌性小鼠,S2给药组小鼠体质量增长量明显高于其他组,S4给药组小鼠体质量增长量明显低于其他组。各给药组小鼠肝、肺、肾组织均出现不同程度病变,以肺组织损伤最严重;各给药组小鼠的肝、肺、肾组织病理学评分均高于对照组,其中在肝组织中S3、S4、S5给药组,在肺组织中S2、S3、S4、S5组,在肾组织中S3、S4给药组的评分显著高于对照组(P<0.05);S4给药组病理学评分均明显高于其他组,其次为S3给药组。结论 不同产地细辛对小鼠的亚急性毒性存在明显差异,其毒性强度可能与药材中黄樟醚含量有关。

【Abstract】 Objective To provide a reference for the study on subacute toxicity and safety of Asari Radix et Rhizoma from different origins.Methods The maximum dosage method was adopted. 60 KM mice were randomly divided into the control group(equal volume of normal saline) and the S1,S2,S3,S4,and S5 administration groups(the origin of medicinal materials was Liaoning,Jilin,Heilongjiang,Hubei and Shandong,respectively),with 10 mice in each group,half male and half female. Mice were intragastrically administered with medicinal solutions or normal saline(intragastrically volume 4 g/kg),twice a day for 14 consecutive days. The general condition,mortality,and changes in body mass of the mice were observed;after 14 days of administration,the main organs(liver,lung,kidney) tissues of mice were taken and stained with hematoxylin-eosin(HE) for pathological morphology observation and histopathology scoring.Results Compared with the control group,mice in each administration group showed varying degrees of toxicity,exhibiting abnormal behaviors such as increased spontaneous activity,irritable behavior,and subsequent deaths,the mortality rate of female mice in each administration group was higher than that of male mice,and the total mortality rates of mice in the S3 and S4 administration groups(40. 00%) were generally higher than those in the other groups;during the administration period,the overall body mass of male mice in all groups was higher than that of female mice. The body mass increase of mice in the S2 administration group was significantly higher than that in other groups,while the body mass increase of mice in the S4 administration group was significantly lower than that in other groups. The liver,lung,and kidney tissues of mice in each administration group exhibited varying degrees of lesions,with lung tissue being the most severely damaged;the histopathology scores of liver,lung,and kidney tissues in each administration group of mice were higher than those in the control group. Among them,compared with the control group,scores in the S3,S4,and S5 administration groups for liver tissue,S2,S3,S4,and S5 administration groups for lung tissue,and S3,S4 administration groups for kidney tissue were significantly higher(P < 0. 05);the histopathology scores in the S4 administration group were significantly higher than those of other groups,followed by the S3 administration group.Conclusion There are significant differences in the subacute toxicity of Asari Radix et Rhizoma from different origins in mice,and the toxicity intensity may be related to the safrole content in the medicinal material.
