分子荧光淬灭在基础实验教学中的探索

【摘要】 提出了一个利用大型仪器瞬态荧光光谱仪改造和提升传统经典荧光分析法的仪器分析实验,以硫酸奎宁的荧光淬灭为研究对象,通过对比分析不同浓度溴化钾(KBr)对硫酸奎宁淬灭状态下的荧光平均寿命衰减、量子产率和紫外吸收光谱的变化来综合判断KBr对硫酸奎宁的淬灭类型。课程内容综合性强、内容丰富,授课形式以线上视频预习与线下实践相结合的方式促进理论知识与实践的融会贯通,有助于学生更全面地掌握瞬态~/稳态的荧光分析技术,认识瞬态荧光光谱仪的结构并了解其功能和应用,同时能开拓学生科研视野、凝练学生的科研思维和锻炼学生的综合科研能力,为提高本科生综合素质和培养卓越工程师打下良好的基础。

【Abstract】 A large instrument transient fluorescence spectrometer was used to transform and improve the traditional fluorescence analysis method. The fluorescence quenching of quinine sulfate was studied. The type of quenching of quinine sulfate by KBr was determined by comparing and analyzing the changes of fluorescence average lifetime decays, quantum yield and ultraviolet absorption spectra of quenched quinine sulfate with different concentrations of potassium bromide. The course content is comprehensive and rich. The teaching form combines online video preview with offline practice to promote the integration of theoretical knowledge and practice, which helps students to master the transient~/steady-state fluorescence analysis technology more comprehensively, understand the structure of the transient fluorescence spectrometer, and understand its function and application. At the same time, it can broaden the students’ scientific research vision, refine their scientific research thinking and exercise their comprehensive scientific research ability, and lay a good foundation for improving the comprehensive quality of undergraduates and training excellent engineers.
