地椒活性成分对HaCaT细胞增殖及迁移的影响

【摘要】 目的 探究不同地椒活性成分(百里香酚、香芹酚、4-异丙基甲苯、β-反式石竹烯和挥发油)对人永生化角质形成细胞(human keratinocyte cell line, HaCaT)增殖及迁移的作用。方法 通过不同地椒活性成分按不同浓度剂量(0.001~0.5 mol/L)给药HaCaT细胞后,利用MTT比色法及细胞划痕实验检测其对细胞增殖和迁移的影响,并利用荧光实时定量PCR技术检测HaCaT细胞中炎症与增殖通路基因表达的变化。结果 与对照组相比,各地椒活性成分对HaCaT细胞增殖和迁移均产生了不同程度的抑制作用,其中香芹酚、β-反式石竹烯和4-异丙基甲苯的效果最为显著。且β-反式石竹烯处理HaCaT细胞后,能显著促进抑炎因子IL-10和IL-34的表达及抑制TNF-α、IFN-γ和NF-κB p65等炎症通路基因的表达,并抑制PI3K/AKT信号通路(phosphatidylinositol 3-kinase/protein kinase b signaling pathway,磷脂酰肌醇3激酶/蛋白激酶B信号通路)基因及促进Notch/HES-1信号通路(notch/hairy/enhancer of split-1 signaling pathway,Notch/HES-1)基因的表达。结论 本研究发现地椒活性成分香芹酚、β-反式石竹烯与4-异丙基甲苯可能通过显著抑制NF-κB p65及其上游IFN-γ与TNF-α基因的表达,并协同抑制HBEGF、EGFR、PI3K、AKT、CREB基因的表达,从而对HaCaT细胞增殖和迁移产生抑制作用。

【Abstract】 Objective To investigate the effects of different active components of Thymus quinquecostatus(thymol, carvacrol, 4-isopropyltoluene, β-caryophyllene, and volatile oil) on the proliferation and migration of HaCaT cells(Human Keratinocyte cell line). Methods HaCaT cells were treated with various active components of thyme at different concentrations(0. 001~0. 5 mol/L). The effects on cell proliferation and migration were assessed using the MTT colorimetric assay and scratch wound healing assay, respectively. Changes in the expression of genes related to inflammation and proliferation pathways were detected by quantitative real-time PCR. Results Compared to the control group, all active components inhibited HaCaT cell proliferation and migration to varying degrees. Carvacrol, β-caryophyllene, and p-cymene exhibited the most significant effects. Furthermore, β-caryophyllene promoted the expression of anti-inflammatory factors IL-10 and IL-34 while simultaneously suppressing the expression of key inflammatory pathway genes, including TNF-α, IFN-γ, and NF-κB p65. Additionally, they inhibited the expression of genes in the PI3K/AKT signaling pathway and promoted the expression of genes in the Notch/HES-1(notch/hairy/enhancer of split-1 signaling pathway) signaling pathway. Conclusion This study demonstrates that the active components of Thymus quinquecostatus-carvacrol, β-caryophyllene, and 4-isopropyltoluene inhibit the proliferation and migration of HaCaT cells. The underlying mechanism likely involves the suppression of NF-κB p65 and its upstream regulators IFN-γ and TNF-α, combined with the coordinated downregulation of HBEGF, EGFR, PI3K, AKT, and CREB gene expression.
