淀粉样前体蛋白及去整合素和金属蛋白酶10在食管鳞状细胞癌中的表达及临床意义
Expression and clinical significance of amyloid precursor protein and a disintegrin and metalloproteinase 10 in esophageal squamous cell carcinoma
【摘要】 目的 探讨淀粉样蛋白前体蛋白(APP)、去整合素和金属蛋白酶10 (ADAM10)在食管鳞状细胞癌(ESCC)癌组织和细胞中的表达及临床意义。方法 采用实时荧光定量反转录聚合酶链式反应和Western blot法分别检测正常人食管黏膜上皮细胞HEEC和人食管鳞状细胞癌细胞系TE-1、KYSE-30、KYSE-150中APP、ADAM10 mRNA及蛋白表达水平。采用免疫组织化学法检测川北医学院附属医院101例ESCC患者切除癌组织和癌旁组织中APP、ADAM10的表达,并分析其临床病理意义。结果 TE-1、KYSE-30、KYSE-150细胞中APP、ADAM10 mRNA和蛋白相对表达量均显著高于HEEC细胞(P<0.05)。免疫组织化学结果显示,APP、ADAM10在ESCC癌组织中的高表达率显著高于癌旁组织(P<0.05),且癌组织中APP与ADAM10的表达呈正相关(r=0.232,P=0.020)。APP表达与患者的年龄、肿瘤分化程度、N分期及淋巴结转移有关(P<0.05),与患者的性别、肿瘤部位、肿瘤直径、T分期、神经侵犯及脉管侵犯均无关(P>0.05)。ADAM10的表达与患者的T分期、N分期、淋巴结转移有关(P<0.05),与患者的性别、年龄、肿瘤部位、肿瘤直径、分化程度、神经侵犯及脉管侵犯均无关(P>0.05)。APP、ADAM10低表达患者的生存率显著高于APP、ADAM10高表达患者(P<0.05)。结论 APP、ADAM10在ESCC细胞及癌组织中均呈高表达,并与ESCC的发生发展有关,检测APP与ADAM10的表达水平对预测患者预后具有一定价值。
【Abstract】 Objective To explore the expression levels and clinical significance of amyloid precursor protein(APP) and a disintegrin and metalloproteinase 10(ADAM10) in esophageal squamous cell carcinoma(ESCC) tissues and cells.Methods Real-time fluorescence quantitative reverse transcription-polymerase chain reaction and Western blot were used to detect the mRNA and protein expression levels of APP and ADAM10 in normal human esophageal mucosal epithelial cells HEEC and in human ESCC cells TE-1,KYSE-30,and KYSE-150.Immunohistochemistry was used to detect the expressions of APP and ADAM10 in cancer and adjacent tissues resected from 101 patients with ESCC at the Affiliated Hospital of North Sichuan Medical College, and their clinical pathological significance was analyzed.Results The relative expression levels of APP and ADAM10 mRNA and protein in TE-1,KYSE-30,and KYSE-150 cells were significantly higher than those in HEEC cells(P<0.05).Immunohistochemical results showed that the high expression rates of APP and ADAM10 in ESCC cancer tissues were significantly higher than those in the adjacent cancer tissues(P<0.05).Furthermore, the expression of APP in cancer tissues was positively correlated with the expression of ADAM10(r=0.232,P=0.020).The expression of APP was associated with patient′s age, tumor differentiation grade, N stage, and lymph node metastasis(P<0.05),but showed no significant correlation with patient′s gender, tumor location, tumor diameter, T stage, nerve invasion, and vascular invasion(P>0.05).The expression of ADAM10 was associated with T stage, N stage, and lymph node metastasis(P<0.05),but showed no significant correlation with patient′s gender, age, tumor location, tumor diameter, differentiation grade, nerve invasion, and vascular invasion(P>0.05).Patients with low APP and ADAM10 expression showed significantly higher survival rates than those with high APP and ADAM10 expression(P<0.05).Conclusion APP and ADAM10 are highly expressed in both ESCC cells and cancer tissues, and are associated with the development and progression of ESCC.Detecting the expression levels of APP and ADAM10 has certain value in predicting patient prognosis.
