川渝苍术黑斑病病原鉴定及其对6种杀菌剂敏感性测定
Identification of Black Spot Disease in Atractylodes lancea from Sichuan and Chongqing and Mensuration of Its Sensitivity to 6 Fungicides
【摘要】 为鉴定引发川渝地区苍术黑斑病的病原,并测定其对常用杀菌剂的敏感性,将四川、重庆苍术种植基地采集的病原样品分离、纯化,进行形态学观察和病原致病性测定,结合病原菌ITS和TEF1-α基因序列进行NCBI比对,构建基因发育树分析确定病原菌种类;选取6种常用的杀菌剂,用菌丝生长速率法测定其对苍术黑斑病病原菌的毒力方程,筛选出高效的抑菌药剂。结果显示:从两地病株中均分离得到了形态相似的菌株;通过ITS序列及TEF1-α序列引物扩增的片段,构建系统发育树并进行同源性比对,综合分析苍术黑斑病病原菌为细极链格孢(Aternaria tenuissima)。杀菌剂毒力测定结果表明:苍术黑斑病病原菌对戊唑醇、苯醚甲环唑、腈菌唑、多菌灵均敏感,其中对多菌灵最敏感,有效抑制中浓度(EC50)仅为0.50μg/mL。
【Abstract】 To identify the pathogens causing black spot disease of Atractylodes lancea in the Sichuan-Chongqing regions and determine its sensitivity to six commonly used fungicides, pathogen samples were collected from A. lancea cultivation bases in Sichuan and Chongqing, were isolated and purified, and subjected to morphological observation and pathogenicity testing. The pathogens were identified by comparing the ITS and TEF1-α gene sequences with NCBI, and a phylogenetic tree was constructed for species determination. Six commonly used fungicide technical materials were selected, and the toxicity equations against the black spot disease pathogen of A. lancea were determined by the mycelial growth rate method to screen out effective bacteriostatic agents. The results showed that similar strains were isolated from diseased plants in both regions. Through the construction of a phylogenetic tree and homology comparison of the ITS and TEF1-α sequences, it was comprehensively analyzed that the pathogen causing black spot disease of Atractylodes was Aternaria tenuissima. The fungicide toxicity test results showed that the pathogen was sensitive to tebuconazole, difenoconazole, myclobutanil, and carbendazim, among which it was most sensitive to carbendazim, with an EC50 value of only 0.50 μg/mL.
