槲皮素对绵羊MⅡ期卵母细胞玻璃化冷冻的影响
The effect of quercetin on vitrification of sheep MⅡ-stage oocytes
【摘要】 旨在研究槲皮素(quercetin,QUE)对绵羊MⅡ期卵母细胞在玻璃化冷冻中的抗氧化应激作用。绵羊卵母细胞体外成熟24 h后,随机分为5组:阴性对照组(新鲜卵母细胞),模型组(冷冻处理,解冻液中不添加QUE),低剂量组(冷冻处理,解冻液中添加2.5μmol/L QUE),中剂量组(冷冻处理,解冻液中添加5μmol/L QUE)和高剂量组(冷冻处理,解冻液中添加10μmol/L QUE)。解冻后,各组卵母细胞均置于添加相应浓度QUE(0、2.5、5、10μmol/L)的体外成熟液中继续培养0.5 h,随后检测各组卵母细胞的存活率、活性氧(ROS)水平、谷胱甘肽(GSH)含量及线粒体膜电位(JC-1)水平。结果:与阴性对照组比,模型组、低剂量组、高剂量组存活率、GSH、JC-1水平显著降低(P<0.05),ROS水平显著升高(P<0.05),但中剂量组ROS水平差异不显著(P>0.05);与模型组相比,低剂量组、中剂量组可显著提高卵母细胞存活率、GSH、JC-1水平(P<0.05),显著降低ROS水平(P<0.05),但高剂量组卵母细胞存活率、GSH、JC-1水平显著降低(P<0.05),ROS水平显著升高(P<0.05)。综上所述,在绵羊MⅡ期卵母细胞玻璃化冷冻过程中添加中剂量的QUE(5μmol/L)可显著减少氧化应激的影响,为提高卵母细胞冷冻保存效率提供研究基础。
【Abstract】 This study aimed to investigate the antioxidant effects of quercetin(QUE) on sheep MII-stage oocytes during vitrification. After in vitro maturation for 24 h,sheep oocytes were randomly divided into five groups: the negative control group(fresh oocytes,without vitrification),the model group(vitrified,with 0 μmol/L QUE added to the vitrification solution),the low-dose group(vitrified,with 2. 5 μmol/L QUE),the medium-dose group(vitrified,with 5 μmol/L QUE),and the high-dose group(vitrified,with 10 μmol/L QUE). After thawing,the oocytes from all groups were cultured in in vitro maturation medium supplemented with corresponding concentrations of QUE(0,2. 5,5,and 10 μmol/L) for 0. 5 h,and then the oocyte survival rate,reactive oxygen species(ROS) levels,glutathione(GSH) content,and mitochondrial membrane potential(JC-1) levels were assessed. The results showed that,compared with the negative control group,the survival rate,GSH content,and JC-1 levels were significantly decreased(P<0. 05),while the ROS levels were significantly increased(P<0. 05) in the model group,the low-dose group,and the high-dose group. However,no significant difference in the ROS levels was observed in the medium-dose group(P>0. 05). Among the vitrified groups,the low-dose and medium-dose groups significantly increased the oocyte survival rate,GSH content,and JC-1 levels(P<0. 05) and significantly decreased the ROS levels(P< 0. 05),compared with the model group. In contrast,the high-dose group showed a significantly decreased survival rate,GSH content,and JC-1 levels(P<0. 05) and significantly increased the ROS levels(P < 0. 05),compared with the model group. In conclusion,the addition of a medium dose of QUE(5μmol/L) during vitrification of sheep MII-stage oocytes significantly alleviated their oxidative stress,which might serve as a basis for improving the efficiency of oocyte cryopreservation.
