基于mRNA技术制备与鉴定牛结节性皮肤病病毒焦磷酸酶多克隆抗体

【摘要】 脱氧鸟苷三磷酸焦磷酸酶(dUTPase)作为核苷酸代谢的关键调控因子,在自然界中广泛分布,多种病毒基因组编码的dUTPase被报道具有独特的生物学功能。本研究旨在探究牛结节性皮肤病病毒(LSDV)编码的dUTPase具体功能。基于LSDV/XJ/201901毒株基因序列,采用mRNA疫苗技术构建脂质纳米颗粒(LNP)包装的LSDV dUTPase mRNA(LNP-dUTPase mRNA),将其作为抗原免疫BALB/c小鼠,制备抗LSDV dUTPase的多克隆抗体。经免疫印迹试验与间接免疫荧光试验验证,所获抗体可特异性识别细胞外源表达蛋白。本研究创新性地通过mRNA免疫技术制备出抗dUTPase蛋白多克隆抗体,为解析LSDV dUTPase的生物学功能提供了关键试验材料。

【Abstract】 Deoxyuridine triphosphate pyrophosphatase(dUTPase),as a key regulatory factor of nucleotide metabolism,is widely distributed in nature. dUTPase encoded by various viral genomes has been reported to have unique biological functions. However,the specific function of the dUTPase encoded by lumpy skin disease virus(LSDV) remains unclear. In this study,based on the gene sequence of the LSDV/XJ/201901 strain,lipid nanoparticle(LNP) packaged LSDV dUTPase mRNA(LNP dUTPase mRNA) was constructed using mRNA vaccine technology as an antigen to immunize BALB/c mice and prepare polyclonal antibodies against LSDV dUTPase. The results were that Western blot and indirect immuno fluorescence assays confirmed that the obtained antibodies could specifically recognize exogenously expressed proteins. This study marked the first successful preparation of polyclonal antibodies against dUTPase protein using mRNA immunization technology,providing key experimental materials for elucidating the biological functions of LSDV dUTPase.
