TA102菌株对纳米农药阿维菌素的吸收特性研究

【摘要】 目的 明确鼠伤寒沙门氏菌(S. typhimurium) TA102菌株对两类纳米农药——活性成分直接纳米化的阿维菌素纳米混悬剂(Aba-NSs)和载体包覆型的阿维菌素纳米囊(Aba-NCs)的吸收特性。方法 以TA102菌株为研究对象,采用透射电子显微镜观察菌株对纳米农药的吸收情况,利用流式细胞术定量分析其摄取水平,并通过高效液相色谱法检测细菌细胞内阿维菌素的含量。结果 透射电子显微镜下观察到Aba-NSs可被细菌直接摄取;流式细胞术进一步证实Aba-NSs处理组的细菌侧向散射信号(SSC)明显增强Aba-NSs,差异有统计学意义(F=749.908,P<0.05);高效液相色谱检测结果发现,Aba-NSs与Aba-NCs处理组细菌均能检测到阿维菌素。结论 在传统Ames试验体系中,试验TA102菌株能直接摄取粒径较小的Aba-NSs,而Aba-NCs虽无法被完整摄取,但其活性成分仍能进入菌株内。从直接遗传毒性的角度分析,Ames试验对纳米农药的评价不受菌株对纳米农药吸收差异的限制。

【Abstract】 Objective To elucidate the absorption characteristics of S.typhimurium TA102 towards two types of nanopesticides:abamectin nano-suspension( Aba-NSs) whose active ingredient is directly nano-sized and abamectin nano-capsules( Aba-NCs) which are carrier-coated. Methods The TA102 strain was treated with the two nano-pesticides( Aba-NSs and Aba-NCs). The uptake of the nanopesticides by bacterial was observed using transmission electron microscopy( TEM). The uptake level was quantified by flow cytometry analyzing the side scatter( SSC) signal,and the intracellular abamectin was detected by high performance liquid chromatography( HPLC).Results Through TEM observation it is confirmed that Aba-NSs could be ingested by bacteria. The result of FCM further confirmed that the SSC of bacteria treated with Aba-NSs was significantly enhanced( F = 749.908,P<0. 05). HPLC detection revealed that abamectin could be detected in the bacterial cells treated with both Aba-NSs and Aba-NCs. Conclusion Under the conditions of the traditional Ames test,the TA102 strain can directly take up the smaller-sized Aba-NSs. Although Aba-NCs cannot be completely internalized,their active ingredients can still enter the bacterial cells. Therefore,from the perspective of direct genotoxicity assessment,the evaluation of nanopesticides using the Ames test is not limited by the absorption characteristics of the strain towards these nanopesticides.
