兰州大尾羊尾脂共轭亚油酸的提取及其体外抗氧化活性研究

【摘要】 兰州大尾羊肉质鲜嫩,具有独特的风味,其尾脂富含共轭亚油酸(Conjugated linoleic acid,CLA)。CLA具有改善心血管健康、增强免疫力及调节代谢等多种生理功能。本研究以兰州大尾羊尾脂为原料,运用尿素包合法提取CLA,以CLA得率及纯度为评价指标,通过单因素实验和Box-Behnken响应面法,优化兰州大尾羊尾脂中CLA的提取工艺参数,并分析其体外抗氧化活性。研究结果显示,尿素包合法提取兰州大尾羊尾脂CLA的最优工艺条件为质量比为1:3.9 g/g、料液比为1:2.2 g/mL、包合温度为46.2℃、包合时间为10 h,此时CLA的纯度为(25.50±0.44)mg/g,得率为(7.25±0.10)mg/g。与低温结晶法提取羊尾脂CLA的纯度((18.62±0.51)mg/g)相比有明显提升。体外抗氧化实验结果显示,当CLA质量浓度为6 mg/mL时,对1,1-二苯基-2-三硝基苯肼(1,1-Diphenyl-2-picrylhydrazyl, DPPH)自由基、2,2’-联氮-双(3-乙基苯并噻唑啉-6-磺酸)(2,2’-Azino-bis(3-ethylbenzothiazoline-6-sulfonic acid),ABTS)阳离子自由基、羟自由基和超氧阴离子自由基的清除率分别为(35.87±0.82)%、(51.68±1.04)%、(86.34±0.68)%和(98.72±0.77)%。本研究为兰州大尾羊尾脂高纯度CLA的工业化提取及功能性产品开发提供一定参考。

【Abstract】 The meat of Lanzhou large-tailed sheep is known for its freshness, tenderness, and unique flavor, while its tail fat is rich in conjugated linoleic acid(CLA). CLA exhibits various physiological functions, including improving cardiovascular health, enhancing immunity, and regulating metabolism. In this study, CLA was extracted from the tail fat of Lanzhou large-tailed sheep using the urea inclusion method. By evaluating the yield and purity of CLA, the extraction process parameters were optimized through single-factor experiments and the Box-Behnken response surface methodology. Additionally, the in vitro antioxidant activity of CLA was analyzed. The results demonstrated that the optimal conditions for extracting CLA from the tail fat of Lanzhou large-tailed sheep via the urea inclusion method were as follows: a mass ratio of 1:3.9 g/g, a material-to-liquid ratio of 1:2.2 g/mL, an inclusion temperature of 46.2 ℃, and an inclusion time of 10 hours. Under these conditions, the purity of CLA reached(25.50±0.44) mg/g, with a yield of(7.25±0.10)mg/g. The purity of CLA extracted from sheep tail fat by low temperature crystallization was(18.62±0.51)mg/g, which was significantly improved. Furthermore, in vitro antioxidant experiments revealed that when the mass concentration of CLA was 6 mg/mL, the scavenging rates for 1,1-diphenyl-2-picrylhydrazyl(DPPH) radical, 2,2’-azino-bis(3-ethylbenzthiazoline-6-sulfonic acid)(ABTS) cation radical, hydroxyl and superoxide anion radical were(35.87±0.82)%,(51.68±1.04)%,(86.34±0.68)%, and(98.72±0.77)%, respectively. These findings provide valuable insights for the industrial optimization of CLA extraction from the tail fat of Lanzhou large-tailed sheep and further studies on its in vitro antioxidant activity.
