超声波辅助可切换DES提取连翘红茶多酚的工艺优化
Optimization of Ultrasound-Assisted Switchable DES Extraction Process of Polyphenols from Forsythia suspensa Red Tea
【摘要】 为探索一种高效、绿色、可持续提取连翘红茶多酚的方法,以连翘红茶为原料,采用超声波辅助可切换低共熔溶剂(deep eutectic solvent,DES)提取多酚,在单因素试验的基础上设计响应面试验优化提取工艺。并利用可切换DES的特性,将用于提取的亲水性DES切换为疏水性DES以实现疏水性DES的回收,被回收的疏水性DES可以切换为亲水性DES继续用于连翘红茶多酚提取。结果表明,可切换DES提取连翘红茶多酚得率高于水或60%乙醇溶液提取,分别高2.65倍、1.31倍。超声波辅助可切换DES提取连翘红茶多酚的最佳工艺条件为料液比1∶40(g/mL)、超声温度70℃、超声时间43 min,在此条件下,连翘红茶多酚得率为(82.590 3±0.377 7)mg/g。可切换DES在重复使用4次后,连翘红茶多酚得率仍较高。
【Abstract】 In order to explore an efficient,green,and sustainable method for extracting polyphenols from Forsythia suspensa red tea,ultrasonic-assisted switchable deep eutectic solvent(DES)was used to extract polyphenols using Forsythia suspensa red tea as raw material. Based on single-factor experiments,response surface methodology was designed to optimize the extraction process. By utilizing the characteristics of switchable DES,the hydrophilic DES used for extraction was switched to a hydrophobic state to realize the recovery of hydrophobic DES,and the recovered hydrophobic DES could be switched back to hydrophilicity for the continuous extraction of Forsythia suspensa red tea polyphenols. The results showed that the extraction yield of Forsythia suspensa red tea polyphenols by switchable DES was higher than that by water or 60% ethanol solution,which was 2.65 times and 1.31 times,respectively. The optimal extraction conditions were obtained as follows:solid-liquid ratio of 1∶40(g/mL),ultrasonic temperature of 70 ℃,and ultrasonic time of 43 min. Under these conditions,the yield of Forsythia suspensa red tea polyphenols was(82.590 3±0.377 7)mg/g. The yield of polyphenols remained high after the switchable DES was reused for four times.
