MALDI-TOF-MS技术鉴定食品真菌的可行性分析
Feasibility Analysis of MALDI-TOF-MS Technology for Identifying Food Fungi
【摘要】 探讨基质辅助激光解析电离飞行时间质谱(MALDI-TOF-MS)技术鉴定食品真菌的应用价值及可行性。使用MALDI-TOF-MS技术鉴定四川省102株不同来源的食品真菌,鉴定结果与传统形态学鉴定方法和测序结果进行比较,评价MALDI-TOF-MS技术对食品真菌的鉴定情况。结果表明:102株食品真菌,用测序方法能100%鉴定到种,形态学方法鉴定到种的准确率为32.35%(33/102), MALDI-TOF-MS技术鉴定到种的准确率为81.37%(83/102),剔除5株未纳入数据库的菌株后, MALDI-TOF-MS技术鉴定到种的准确率为85.57%(83/97)。对于青霉属、曲霉属、毛霉属、茎点霉属、镰刀菌属等常见食品真菌, MALDI-TOF-MS技术鉴定到种的准确率均可达100%。MALDI-TOF-MS技术适用于食品真菌的快速鉴定。
【Abstract】 Explore the application value and feasibility of matrix assisted laser desorption ionization time-of-flight mass spectrometry(MALDI-TOF-MS) technology in food fungal identification. Identify 102 food fungi from different sources in Sichuan Province using MALDI-TOF-MS technology. The identification results were compared with traditional morphological identification methods and sequencing results to evaluate the identification of food fungi using MALDI-TOFMS technology. The results showed that for 102 strains of food fungi, the species could be identified 100% using sequencing methods, the identification accuracy rate of morphological method was 32.35%(33/102), and the identification accuracy rate of MALDI-TOF-MS technology was 81.37%(83/102). After excluding five strains that were not included in the database, the identification accuracy rate of MALDI-TOF-MS technology was 85.57%(83/97). For food fungi, such as Penicillium, Aspergillus, Mucor, Rhizopus and Fusarium, the accuracy of MALDI-TOF-MS technology in identifying species could reach 100%. MALDI-TOF-MS technology is suitable for rapid identification of food fungi.
