玉蚕颗粒的制备工艺优化研究
Research on optimizing preparation process of Yucan Granules
【摘要】 目的:对玉蚕颗粒的制备工艺进行优化研究。方法:采用单因素试验法,系统考察黄芪、川芎、玉米须、蚕茧壳的浸泡时间、加水量、提取时间、提取次数、过滤材质与目数、浓缩温度及干燥方式等因素对提取效果的影响。首次将蚕茧壳单煎,与合煎工艺比较,进行药效学研究。结果:确定提取工艺为黄芪、川芎、玉米须合煎,加12倍量水,浸泡90 min,提取2次,采用300目滤布单层过滤;蚕茧壳单煎,加16倍水,浸泡60 min,煎煮3次,每次1 h,以100目滤布单层趁热过滤,合并滤液。以80℃为减压浓缩温度,进行喷雾干燥保存。所制得的玉蚕颗粒在成分保留及药效学评价方面,均具有良好应用价值。结论:优化后的制备工艺操作简便、流程简洁,可最大程度保留药物的有效成分;该工艺不仅能充分挖掘蚕茧壳的药用价值,还可实现蚕丝资源的循环利用,具备良好的经济与应用价值。采用该工艺制得的玉蚕颗粒具有补肾活血、清化湿瘀的功效,对改善水肿、缓解肾脏相关病症具有显著效果。
【Abstract】 Objective: To optimize the preparation process of Yucan Granules. Methods: A single-factor experimental design was conducted to systematically investigate the effects of various factors on the extraction efficacy, including the soaking time, water volume, extraction duration, extraction cycles, filtration material and mesh size, concentration temperature, and drying method for Astragali Radix, Chuanxiong Rhizoma, Maydis Stigma(Corn Silk), and Silkworm Cocoon Shells. For the first time, a pharmacodynamic study was conducted to compare the separate decoction of Silkworm Cocoon Shells with the combined decoction of all herbs. Results: The optimal extraction process was determined as follows: Astragali Radix, Chuanxiong Rhizoma, and Maydis Stigma are decocted together with 12 times the amount of water, soaked for 90 minutes, and extracted twice, followed by single-layer filtration through 300-mesh filter cloth. Silkworm Cocoon Shells were decocted separately with 16 times the amount of water, soaked for 60 min, and extracted three times, each for 1 h, followed by immediate hot filtration through single-layer 100-mesh filter cloth. The combined filtrates were concentrated under reduced pressure at 80℃ and then subjected to spray drying for preservation. The prepared Yucan Granules exhibit excellent application value in terms of both active ingredient retention and pharmacodynamic evaluation. Conclusion: The optimized preparation process is simple to operate with a streamlined procedure, maximizing the retention of the active components in the herbs. This process not only fully exploits the medicinal value of Silkworm Cocoon Shells but also enables the recycling of silk resources, offering significant economic and application value. The Yucan Granules produced with this process exhibit effects in tonifying the kidney and activating blood, as well as resolving dampness and stasis, showing significant efficacy in alleviating edema and improving kidney-related disorders.
