基于网络药理学探讨石菖蒲-川芎防治血管性痴呆作用机制
Approach to the mechanism of effect of drug pair rhizoma acori tatarinowii-rhizoma Chuanxiong on prevention and treatment of vascular dementia,based on network pharmacology
【摘要】 目的:基于网络药理学方法探讨石菖蒲、川芎药对防治血管性痴呆的分子作用机制。方法:利用中药系统药理学数据库分析平台(TCMSP数据库)搜集石菖蒲、川芎的主要活性成分及其潜在作用靶点;通过GeneCard、OMIM等数据库筛选出血管性痴呆的疾病靶点,将其与活性成分靶点取交集即为药物和疾病的共同作用靶点;基于STRING数据库,构建石菖蒲、川芎药对防治血管性痴呆的蛋白互作网络;利用Cytoscape3.9.1软件构建成分疾病靶点的可视化网络分析图;利用DAVID数据库对共同作用靶点进行GO功能和KEGG信号通路富集分析,预测其作用机制;采用AutoDock等软件对核心靶点和其作用的活性成分进行分子对接验证。结果:从TCMSP数据库中检索到石菖蒲、川芎药对的7个有效活性成分以及对应的388个成分靶点,与VD的1 395个靶点取交集后得到141个共同作用靶点,核心靶点涉及MMP2、NR3C1、KDR、STAT3、EGFR、CASP3、GSK3B等;GO富集分析得到生物学过程相关条目326条、分子功能相关条目63条以及细胞组分相关条目79条(P<0.01);KEGG通路富集分析得到127条信号通路(P<0.01);分子对接结果显示活性成分与核心靶点均有较好的结合活性。结论:石菖蒲、川芎药对防治血管性痴呆具有多成分、多靶点、多通路的作用特点,其作用机制可能与表皮生长因子受体信号通路、磷脂酰肌醇3-激酶/蛋白激酶B信号通路、Ras-同源蛋白1信号通路、缺氧诱导因子-1信号通路、脂质和动脉粥样硬化通路等有关。
【Abstract】 Objective:To approach the mechanism of molecular effect of drug pair rhizoma acori tatarinowiirhizoma Chuanxiong on prevention and treatment of vascular dementia(VD)based on network pharmacology.Methods:The main active components and potential drug targets of rhizoma acori tatarinowii and phizoma Chuanxiong were collected by Traditional Chinese Medicine Systems Pharmacology Database and Analysis Platform(TCMSP).The disease targets of vascular dementia were screened out by databases such as GeneCard,OMIM,etc.,and the intersections of these disease targets with those of active components were just the common action target of drugs and disease.A protein interaction network of drug pair rhizoma acori tatarinowii-rhizoma Chuanxiong for preventing and treating vascular dementia was constructed by STRING database.A visual network analysis diagram for targets of components and disease was constructed by Cytoscape3.9.1 software.The enrichment analysis of GO function and KEGG signal pathway of common action targets was conducted by DAVID database,and the mechanism of effect of these targets was predicted.The molecular docking of core targets with effective active components of drug pair was verified by softwares such as AutoDock,etc.Results:7 effective active components and 388 corresponding component targets of drug pair rhizoma acori tatarinowiirhizoma Chuanxiong were retrieved out from TCMSP database;141 common action targets were obtained after intersecting with 1 395 targets of VD;the core targets involved MMP2,NR3C1,KDR,STAT3,EGFR,CASP3,GSK3B,etc.326 entries related to biological process,63 entries related to molecular function and 79 entries related to cell components were obtained by GO enrichment analysis(P﹤0.01).127 signal pathways were obtained by KEGG pathway enrichment analysis(P ﹤ 0.01).The results of molecular docking showed that both the active components and the core targets had good binding activity.Conclusion:The drug pair rhizoma acori tatarinowii-rhizoma Chuanxiong has the characteristic of effect of multi-components,multi-targets and multi-pathways on prevention and treatment of vascular dementia,and its effect mechanisms may be related to following signal pathways such as epidermal growth factor receptor signal pathway,phosphatidylinositol 3-kinase/protein kinase B signal pathway,ras-association proximate 1 signal pathway,hypoxia-inducible factor-1 signal pathway,lipid and atherosclerosis pathway,etc.
