甘草减轻黄药子致小鼠肝毒性机制研究
Approach to mechanism of radix glycyrrhizae reducing rhizoma dioscoreae bulbiferae-induced hepatotoxicity in mice
【摘要】 目的:通过microRNA测序,从基因测序水平探究甘草配伍黄药子减轻肝毒性的实验机理。方法:将实验用小鼠随机分为黄药子组、配伍组、空白对照组,每组各10只,分别连续灌胃14天后,HE染色进行肝组织形态观察,同时采用转录组学技术测序肝组织microRNA基因表达谱,应用软件分析表达谱提示存在差异表达的基因。结果:共筛选出具有差异表达的microRNA共93个,其中黄药子组有57个microRNA显著下调,显著上调的有36个。结论:甘草可能通过膜电位调节、突触后特化结构、离子信号活性等生物功能;以及轴突导向和Ras信号通路、细胞信号转导通路(MAPK)等多通路调节降低黄药子毒性导致肝损伤。
【Abstract】 Objective:To approach the experimental mechanism of compatibility of radix glycyrrhizae with rhizoma dioscoreae bulbiferae alleviating hepatotoxicity based on the level of gene sequencing,by microRNA sequencing.Methods:The experimental mice were randomly divided into rhizoma dioscoreae bulbiferae group,compatibility group and blank control group,10 mice in each group.After 14 days of continuous gavage,the morphology of liver tissue was observed by HE staining.microRNA gene expression profile of liver tissue was sequenced by transcriptomics technology at the same time.It was analyzed by software that the expression profile suggested that there were differentially expressed genes.Results:93 microRNAs with differential expression were screened out in total.Among them,57 microRNAs were significantly down-regulated in rhizoma dioscoreae bulbiferae group,while 36 other were significantly up-regulated.Conclusion:Radix glycyrrhizae can reduce liver impairment caused by toxicity of rhizoma dioscoreae bulbiferae,which may be by biological functions such as membrane potential regulation,postsynaptic specialized structure and ion signal activity,etc.as well as by multi-pathway regulation such as axonal guidance and Ras signal pathway,cell signal transduction pathway(MAPK),etc.
