铜纳米簇比率荧光探针超灵敏检测尿液中左氧氟沙星

【摘要】 采用水热法合成苯丙氨酸模板化铜纳米簇(Phe-CuNCs),通过Phe-CuNCs与罗丹明6G(Rh6G)之间的静电相互作用,构建双发射比率荧光探针。该探针在310 nm激发下,于420 nm和553 nm处出现两个分离良好的发射峰,可在pH=7.0条件下实现人尿液样品中左氧氟沙星(LEV)浓度的测定。为探索Phe-CuNCs/Rh6G对LEV的选择性,除LEV外,还包括3种抗生素的荧光研究,分别为阿奇霉素(AZ)、红霉素(ER)、头孢泊肟(CD),它们与LEV具有结构相似性。LEV浓度在0~5??mol/L范围内连续添加目标物后,得回归方程F1/F2=0.010 4CLEV+1.90,以信噪比S/N=3计算,检测限为0.03μmol/L。在人尿液样品中的实际应用结果显示,该探针的回收率为84.00%~102.00%,具备良好的实用性。

【Abstract】 Phenylalanine-templated copper nanoclusters(Phe-CuNCs) were synthesized via a hydrothermal method. A dual-emission ratiometric fluorescent probe was constructed based on the electrostatic interaction between Phe-CuNCs and Rhodamine 6G(Rh6G). Under the excitation wavelength of 310 nm, the probe exhibited two well-resolved emission peaks at 420 nm and 553 nm, which could be applied to the determination of levofloxacin(LEV) in human urine samples at pH=7.0. To explore the selectivity of Phe-CuNCs/Rh6G toward LEV, fluorescence investigations were also conducted on three other antibiotics besides LEV, namely azithromycin(AZ), erythromycin(ER) and cefpodoxime(CD). With the continuous addition of LEV within the concentration range of 0-5 ??mol/L, the regression equation was fitted as F1/F2=0.010 4CLEV+1.90. The limit of detection was calculated to be 0.03 ??mol/L at a signal-to-noise ratio of S/N=3. The practical application in human urine samples presented satisfactory recoveries ranging from 84.00% to 102.00%, demonstrating favorable practical applicability of the probe.
